Evaluation of fixative solutions for ultrastructural analysis of brown spider Loxosceles intermedia (araneae: sicariidae) tissues

In view of the widely varying compositions of fixative solutions used for studying spiders, five different fixative formulas were tested for fixing male brown-spider (Loxosceles intermedia) gonad tissues. The brown spider represents a public health problem in Curitiba (Paraná State, Brazil). Morphological study of its gonads may aid in understanding the reproductive strategies of this species, and possibly in developing a reproduction control program. The fixatives tested contained glutaraldehyde alone or combined with paraformaldehyde, and the buffers cacodylate or phosphate, with or without the addition of sucrose or sodium chloride as osmolytes. Those containing 2.5% glutaraldehyde and 2% paraformaldehyde in 100 mM phosphate buffer with 200 mM sucrose, or in 200 mM sodium cacodylate, satisfactorily preserved mitochondria, the Golgi apparatus, and the membranes in general. These formulas were nearly isosmotic (439 mOsm/kg H2O and 455 mOsm/kg H2O respectively) to brown spider hemolymph (478 mOsm/kg H2O). With respective to the fixative agents, a glutaraldehyde-paraformaldehyde combination resulted in optimal fixation of Loxosceles intermedia cells. For other species of spiders, hemolymph osmolality should be considered, but the fixative formulas cited above would also probably yield good results.

Parasite fauna monitoring of matrinxã Brycon amazonicus (Spix & Agassiz, 1829) raised in an intensive husbandry system in a stream channel in the state of Amazonas, Brazil

The matrinxã, Brycon amazonicus, is one of the main species being used for aquaculture and fishery activities in the Amazonian region, yet little is known about the parasites that cause its diseases. The present study monitored parasite fauna infesting an intensive husbandry system in a stream channel. The collections were carried out monthly from September, 1997 to January, 1998. Water quality parameters, fish removal procedures, as well as biometry and anaesthetic data were observed in order to monitor the management conditions. Of the 80 sampled fish, 71 (89%) were infested with four Monogenoidea species (Anacanthorus spiralocirrus, Jainus amazonensis, Tereancistrum kerri and Trinibaculum brazilensis), 12 (15%) one of Nematoda Spirocamallanus inopinatus, and three (3.75%) with the Copepoda Ergasilus bryconis. Monogenoidea presented the highest prevalence and intensity of infestation. The Kruskall-Wallis non-parametric variance analysis showed a significant difference (P < 0.05) in the monthly parasite indexes of A. spiralocirrus, J. amazonensis and T. brazilensis, however no significant difference (P > 0.05) was detected for T. kerri. Evaluation of the management conditions pointed out that monthly capture anaesthesia and hauling procedures may have been responsible for some of the clinical signals such as loss of scales, erosion and no pigments on the caudal fin and cornea opacity, which were the most frequently found injuries in husbandry.

The small-scale urban reservoir fisheries of Lago Paranoá, Brasília, DF, Brazil

In many cases in large urban centers, which have appropriate waterbodies, small-scale fisheries are the only source of cheap protein for the poor. In Lago Paranoá, located in Brasília, the capital city of Brazil, fishing was studied by conducting interviews with 53 fishers filling in logbooks from March, 1999 to March, 2000 in three fishing communities. The fishers come from the poorest towns around Brasília, known as satellite-towns. They have been living there on average for 21.7 years (s = 9.6 years), their families have 4.9 members (s = 3.6) on average and 44.2% do not have a basic education. However, such characteristics are similar to the socioeconomic indices of the metropolis where they live. In spite of being illegal between 1966 and 2000, fishing generated an average monthly income of U$ 239.00 (s = U$ 171.77). The Nile Tilapia Oreocrhromis niloticus is the main captured species (85% of a total number of landings in weight of 62.5 t.). Fishing is carried out in rowing boats, individually or in pairs. The fishing equipment used are gillnets and castnets. Gillnets were used actively, whereby the surface of the water is beaten with a stick to drive Tilapias towards nets as they have the ability to swim backwards. This fishing strategy was used in 64.7% of the fisheries, followed by castnets (31.1%) and by gillnets which were used less (4.2%). The fish is sold directly in the streets and fairs of the satellite-towns to middlemen or to bar owners. Three communities have different strategies in terms of fishing equipments, fishing spots and commercialization. Consequently, there are statistically significant differences in relation to the monthly income for each one of these communities.

Anthropic effects on the fish community of Ribeirão Claro, Rio Claro, SP, Brazil

The effect of anthropic alterations such as drain discharge on a fish community was studied in the Ribeirão Claro River, municipality of Rio Claro, State of São Paulo, southeastern Brazil. Samples were made monthly in three different points along the river (headwaters, mid course, and confluence) between December, 2003 and March, 2004, which is the reproductive period for the majority of the species. The fish community of the Ribeirão Claro River showed a fair composition and diversity, with species rarely observed in studies made in the region, such as Paravandellia oxyfera and Callichthys callichthys. Indices of diversity and equitability showed different results when weight or the number of individuals were considered. Moreover, these indices did not reveal the typical increase in diversity from the headwaters toward the confluence, nor the loss of richness in the part altered by drain discharge. An interpretation should be made with caution, taking into account the diverse factors included in the computation. Similar to the diversity indices, the Morisita-Horn similarity index did not reveal a great difference in the fish community of the confluence of the river, mainly in relation to its mid course. The decline of species richness and trophic composition alteration in the disturbed part is clear, which shows a great dominance of piscivorous species.

Are there co-occurrence patterns that structure snake communities in Central Brazil?

The main factors that structure Neotropical animal communities have been the subject of discussion in ecology communities. We used a set of null models to investigate the existence of structure in snake communities from the Cerrado in Central Brazil in relation to the co-occurrence of species and guilds concerning specific resources. We used fragments (conservation units) inside the Distrito Federal and neighbor municipalities. In spite of recent human colonization in the region from the end of the 1950’s, intense habitat modification and fragmentation has taken place. Sixty three snake species are present in the Distrito Federal. Co-occurrence analysis of species and guilds associated to snake diets and habitats suggested a lack of organization. The homogeneity of habitats in Central Brazil and the minor importance of ecological effects can lead to random arrangement.

Niche differences between two sympatric whiptail lizards (Cnemidophorus abaetensis and C. ocellifer, Teiidae) in the restinga habitat of northeastern Brazil

Differences among sympatric lizard species usually result from differences in the use of three resources: space, time and food or some combination of these three. However, differences in resource utilization among sympatric species may simply reflect their specific ecological needs rather than competitive pressures. In this study, we analyzed the temporal, spatial and food niche of two congeneric teiids (Cnemidophorus abaetensis and C. ocellifer) living sympatrically in the "restinga" habitat of Abaeté in the Salvador Municipality, Bahia State, Brazil to assess the degree of niche differentiation among them. The whiptail species overlapped considerably in an hourly activity (Ojk = 0.93), in microhabitat use (Ojk = 0.97) and in the prey items consumed (Ojk = 0.89). Differences in amount of vegetation in the microhabitats used by both lizard species may have contributed to differences in the activity period and in the distribution of the main prey eaten by these lizards which may, in turn, facilitate their coexistence in Abaeté. Although sympatric C. ocellifer and C. abaetensis in Abaeté differed only slightly in their use of microhabitats, period of activity and diet, the most important niche dimension segregating the two species seemed to be the food niche.

Responses of Chironomidae (Diptera; Insecta) to the exclusion of shrimps and Ephemeroptera in a coastal forest stream, Rio de Janeiro, Brazil

In a benthic community on a continuous flat granite substrate in a third-order coastal forest stream, the dominant chironomid (Cricotopus) increased in number when shrimps (Macrobrachium olfersi and Potimirim glabra) and baetid ephemeropterans were excluded by electricity. The response appeared to be mediated by an increase in periphyton and sediments, rather than a reduction of direct predation or interference. Chironomids, periphyton and sediments decreased significantly compared to the control when shrimps only were excluded. Baetid ephemeropteran appeared to be the most important determinants of periphyton and sediment mass; the density of chironomids appeared to follow the quantity of periphyton and sediments.

Abundance and diversity of phytoplankton in the Paraná River (Argentina) 220 km downstream of the Yacyretá reservoir

Patterns in the temporal composition, abundance and diversity of the phytoplankton community of the Paraná river prior to and after the initial filling phase of the Yacyretá reservoir are analyzed. The study site is located 220 km downstream from the Yacyretá reservoir and 30 km downstream from the confluence with the Paraguay river. Because both rivers remain separate and unmixed at the study site, we compared the possible effects of the impoundment on both river banks (left and right banks) in hydrological periods with similar duration and magnitude of the low and high water phases. Physical and chemical conditions measured on the right bank (water from the Paraguay river) were similar at both periods (pre and post-impoundment) whereas conductivity, pH and orthophosphate concentration increased on the left bank (water from the High Paraná river and Yacyretá reservoir) after the impoundment. Changes in phytoplankton density and diversity were observed only in samples collected from water flowing from the reservoir (left bank). The density of Chlorophyceae (Chloromonas acidophila, Chlamydomonas leptobasis, Choricystis minor, Chlorella vulgaris, Scenedesmus ecornis, Monoraphidium minutum, M. contortum and M. pusillum) and Cryptophyceae (Rhodomonas minuta, Cryptomonas marssonii and C. ovata) increased while Cyanophyceae (Cylindrospermopsis raciborskii, Raphidiopsis mediterranea and Planktolyngbya subtilis) and Bacillariophyceae (Aulacoseira granulata and its bioforms) decreased compared to previous studies conducted on the left bank of the Paraná river. Phytoplankton collected from the right bank of the river did not differ in pre and post- impoundment samples because they originate from the Paraguay river, which remains relatively unaffected by human activities.

Food partitioning between breeding White-tailed Kites (Elanus leucurus; Aves; Accipitridae) and Barn Owls (Tyto alba; Aves; Tytonidae) in southern Brazil

I examined the diet of breeding White-tailed Kites (Elanus leucurus; Aves; Accipitridae) and Barn Owls (Tyto alba; Aves; Tytonidae) in an agrarian area of southern Brazil by analyzing regurgitated prey remains. The objective was to evaluate how these raptors, which differ markedly in their hunting activity periods (owls are nocturnal and kites diurnal), share their mammalian food component. 2,087 prey consumed by Barn Owls and 1,276 by White-tailed Kites were identified. They presented a high overlap of food-niches (Pianka’s index was 0.98). Based on the daily activity period of their main small mammal prey, a lower overlap would be expected. The crepuscular/nocturnal Mus musculus was the main prey for the diet of breeding Barn Owls (81%) and White-tailed Kites (63%). This small exotic rodent provided 63% of the small mammal biomass ingested by owls and 44% by kites. Larger native small mammals were also considered important for the diet of kites, mainly because of their biomass contribution. Although these raptors differ markedly in their hunting activity periods, Barn Owls and White-tailed Kites are very similar predators in southern Brazil, overlapping their diets.

Sewage input effects on the macroinvertebrate community associated to Typha domingensis Pers in a coastal lagoon in southeastern Brazil

This study was carried out at Imboassica Lagoon, located in an urban zone in the municipality of Macaé, Rio de Janeiro state, Brazil. This lagoon has been subject to anthropogenic impacts due to the increasing city population, such as the input of sewage. Areas of variable degree of anthropogenic influence in the lagoon were compared regarding the structure of the macroinvertebrate community associated to Typha domingensis leaves. For sampling, we used 35 x 20 cm net plastic bags, with 6.8 mm mesh containing T. domingensis leaves for colonization. Two different sampling stations were selected: station A, under direct input of sewage; and station B with lesser sewage influence. The bags were removed after 20, 40 and 75 days of colonization. For each sample the Shannon-Wiever Diversity, Pielou Evenness, Jaccard Similarity Indices, Correspondence Analysis and taxonomic richness were calculated. A total of 31,874 individuals were sampled, belonging to 34 taxa. The main taxonomical groups were: Oligochaeta (41%), Chironomidae (40%), Ancylidae (4.6%), Polymitarcyidae (4%) and Thiaridae (3%). At station A, the taxonomic richness, the Evenness and Diversity values were lower than in station B. On the other hand, the total density was three times higher in station A than in B. It was already possible to discriminate the community structure of each sampling station in the first sampling. Trichoptera and Ephemeroptera were the main exclusive groups of station B and are considered good water quality indicators due to their high sensibility to contamination. The major contribution to discriminate between the macroinvertebrate communities of the two sample stations came from Chironomidae, Oligochaeta and Ephemeroptera.

Sustainable harvest of two large predatory catfish in the Cuiabá river basin, northern Pantanal, Brazil

Fishery is an important economic activity in the Pantanal. Among the region’s species, the Pimelodidae catfish stands out as an important part of the annual catch. This study assesses the structure, exploitation and stock management of Hemisorubim platyrhynchos and Sorubim cf. lima, the sixth and seventh largest Pimelodidae of the Pantanal. The analysis is based on fish caught by commercial fishing in the Cuiabá river and landed at the "Antônio Moysés Nadaf" Market in the Cuiabá city, Mato Grosso state, Brazil. The findings indicate that commercial fishing activities target several fish cohorts and that usually only individuals above mean length at first maturation are caught. Estimates of the instantaneous mortality coefficient show that the current fishing mortality is low. Simulations of relative yield-per-recruit model demonstrate that the current yield of two species could be greater if the fishery effort were increased, indicating that the stocks are underexploited. However, an increase in current fishery efforts should be viewed with caution, since the stock-recruitment relationship for the species is unknown. The results indicate that the current harvest of two species in the Cuiabá River Basin is sustainable.

Performance consequences of food mixing in two passion vine leaf-footed bugs, Holymenia clavigera (Herbst, 1784) and Anisoscelis foliacea marginella (Dallas, 1852) (Hemiptera; Coreidae)

Holymenia clavigera (Herbst) and Anisoscelis foliacea marginella (Dallas) (Hemiptera: Coreidae: Anisoscelini) are distributed in southern Brazil and use various passion vine species (Passifloraceae) as host-plants. Preliminary observations indicate a high coexistence of these species in terms of host-plant use; in addition, there is a strong similarity regarding egg and nymph morphology. In this study, the most suitable feeding sites for nymph performance on wild (Passiflora suberosa Linnaeus and Passiflora misera Humbold, Bonpland et Kunth) and cultivated (Passiflora edulis Sims) hosts were determined by rearing them on each host and on the combination of hosts. Performance was determined by evaluating nymph development and survivorship, and adult size at emergence. Plant parts used were also recorded. For both species, P. suberosa was the most suitable host plant. First instar nymphs of both species fed on terminal buds more frequently when compared to other plant parts. Second instar nymphs switched to green fruits, whose behavior was more pronounced for H. clavigera. Thus, H. clavigera and A. foliacea marginella immatures are extremely similar in terms of host-plant use and consequences for performance, in addition to their morphological similarity. We suggest that these coreids may have evolved through several processes, including parsimony between the immature stages after speciation, evolutionary convergence, mimicry or genetic drift.

Thermolysin in human cultured keratinocyte isolation

BACKGROUND: When treating extensively burned patients using cultured epidermal sheets, the main problem is the time required for its production. Conventional keratinocyte isolation is usually done using Trypsin. We used a modification of the conventional isolation method in order to improve this process and increase the number of colonies from the isolated epidermal cell population. PURPOSE: To compare the action of trypsin and thermolysin in the keratinocyte isolation using newborn foreskin. METHODS: This method used thermolysin as it selectively digests the dermo-epidermal junction. After dermis separation, the epidermis was digested by trypsin in order to obtain a cell suspension. RESULTS: Compared to the conventional procedure, these experiments demonstrated that in the thermolysin group, the epidermis was easily detached from the dermis, there was no fibroblast contamination and there were a larger number of keratinocyte colonies which had a significant statistical difference. CONCLUSION: The number of colonies in the thermolysin group was significantly greater than in the trypsin group.

Effect of zinc and benzene on respiration and excretion of mussel larvae (Perna perna) (Linnaeus, 1758) (Mollusca; Bivalvia)

The presence of pollutants in the ocean may affect different physiological parameters of animals. Oxygen consumption and ammonia excretion were evaluated in D-shaped larvae of mussels (Perna perna) exposed to zinc sulphate (ZnSO4) and benzene (C6H6). When compared to the control group, both pollutants presented a significant reduction in oxygen consumption. A reduction in the ammonia excretion was also observed, both for ZnSO4 and C6H6 and also in the oxygen consumption. The results indicate that anaerobic metabolism may occur at the beginning of P. perna mussels development, as observed in veliger larvae. The O:N ratio under experimental conditions showed low values indicating that catabolism in veliger larvae was predominantly proteic.

Alternative methodology for isolation of biosurfactant-producing bacteria

Wide biosurfactant application on biorremediation is limited by its high production cost. The search for cheaper biossurfactant production alternatives has guided our study. The use of selective media containing sucrose (10 g.L-1) and Arabian Light oil (2 g.L-1) as carbon sources showed to be effective to screen and maintain biosurfactant-producing consortia isolated from mangrove hydrocarbon-contaminated sediment. The biosurfactant production was assayed by kerosene, gasoline and Arabian Light Emulsification activity and the bacterial growth curve was determined by bacterial quantification. The parameters analyzed for biosurfactant production were the growth curve, salinity concentration, flask shape and oxygenation. All bacteria consortia screened were able to emulsify the petroleum derivatives tested. Biosurfactant production increased according to the incubation time; however the type of emulsification (non-aqueous phase or aqueous phase) did not change with time but with the compound tested. The methodology was able to isolate biosurfactant-producing consortia from superficial mangrove sediment contaminated by petroleum hydrocarbons and was recommended for selection of biosurfactant producing bacteria in tropical countries with low financial resources.

Diet of Physalaemus cf. cicada (Leptodactylidae) and Bufo granulosus (Bufonidae) in a semideciduous forest

We determined the diet of the two most abundant anuran species which occur in the litter of a semideciduous forest (Lençóis, Bahia, Brazil), Physalaemus cf. cicada and Bufo granulosus in the dry and rainy seasons. Pitfall traps were used to collect anuran and invertebrate fauna, which showed the availability of prey in the environment. Physalaemus cf. cicada was present in both seasons and Bufo granulosus only in the rainy season. Both species fed mainly on Isoptera and Formicidae. However, there is a difference between the rainy and dry seasons concerning the diet of P. cf. cicada. During the rainy season P. cf. cicada consumed less Isoptera and more Formicidae than in the dry season. In the volumetric sense, Orthoptera was the most important alimentary category for P. cf. cicada and B. granulosus. The Jacobs’ electivity index indicated that Physalaemus cf. cicada and Bufo granulosus were specialists in Isoptera.
