O Papel da metformina no controle do estresse oxidativo em músculo de ratos diabéticos

CHAPTER II: Introduction and Objectives: Metformin can act in muscle inhibiting the complex I of the electron transport chain and activating a signaling cascade for glut-4 translocation to the membrane cell. Our hypothesis is that this inhibition can decrease mitochondrial reactive oxigen species (ROS), minimizing cell damage caused by oxidative stress. Thus, the aim of this study was to evaluate the activity and expression of oxidative stress enzymes, total antioxidant status and lipid peroxidation in gastrocnemius muscle of rat with induced diabetic Mellitus and treated with metformin. Materials and Methods: The DM was induced in Wistar rats divided into four groups: untreated, treated with insulin, insulin plus metformin, and metformin. A non-diabetic group was used as control. The gastrocnemius muscle was collected and homogenized to evaluate the following parameters: superoxide dismutase (SOD) and catalase (activity and immunoblotting), glutathione peroxidase, glutathione reductase and glucose-6-phosphate dehydrogenase (activity), reduced glutathione, lipid peroxidation and total antioxidant capacity were measured. Results and conclusions: he group treated with metformin showed prevention in increased activity and expression of SOD and catalase, and kept the total antioxidant capacity and G6PDH activity when compared to the untreated group. These results seem to be related to lower lipid peroxidation. Therefore, this study suggest that metformin may is involved in the regulation of the oxidative stress in muscle cells decreasing lipid peroxidation which could improve glucose uptake.

Pneumonia em pacientes sob ventilação mecânica por Staphylococcus aureus resistentes à Meticilina, internados em UTI de adultos : aspectos microbiológicos, clínicos e epidemiológicos

Objectives: Investigate the participation of the S. aureus, as etiological agent of the pneumonias associated to the mechanical ventilation (PAVs) and evaluate the colonization as risk factor of PAV and the prognostic of the treatment when correct or incorrect. Material and Methods: The study was prospective, longitudinal, performed from March/2005 to February/2006, including 500 patients admitted in the UTI - adult, with 15 beds of the Hospital of Clinics of the Federal University of Uberlândia. All the patients were submitted to the colonization research by S. aureus in different sites. The PAVs were defined based on clinical, radiological criteria and microbiological count 106 UFC/ml in the tracheal aspirate. Results: The S. aureus was the most frequent agent in the PAVs. The participation of samples of MRSA was inferior (42,1%) to the one observed by MSSA (57,9%). In the patients with PAV by S. aureus, a most frequent colonization was observed by MRSA. The samples of MRSA behaved as multi resistant and the ones resistant to clindamycin, showed, in the majority, as from the constituent phenotype (60%). In PAVs by S. aureus, the mortality rate was high in the subgroup MRSA (37,5%). Conclusion: The more and more frequent presence of S. aureus in PAVs has been becoming a serious problem, raising the necessity of a reflection about the need of the introduction the de-escalonation of antibiotics regimens.

The biological organization complexes, which comprise proteins, membranes, chromosomes, organelles, cells and tissues, have to keep their stabilities against the deleterious action of factors as variations in temperature, pH and concentration of solutes. Among the protective mechanisms is the stabilization promoted by the action of solutes generally called as osmolytes. Our aim here is understand the origin of such stabilizing action. Thus, we tried to give emphasis in the structural aspects of the membranes that could constitute a target of the osmostabilizing action of the cosmotropic solutes. Since this action has been more profoundly studied in terms of the stabilization of proteins, we also presented the theory more largely commented about the protein stabilization by osmolytes. We believe that the same thermodynamic paradigms used in the study of the action of osmolytes on proteins would bring satisfactory answers about the intimate nature of the osmostabilization of membranes.
