Transfer cells in trichomatous nectary in Adenocalymma magnificum (Bignoniaceae)
ABSTRACT Transfer cells are specialized cells that play an important role where there are high energy costs due to facilitation of transmembrane flow of solutes. This study aimed to investigate the ontogenesis, histochemistry and ultrastructure of glandular trichomes focusing on stalk cells and their possible transfer function. Samples of prophylls of axillary buds, calyces of flowers in anthesis, and flower buds in different stages of development were collected, fixed and processed according to the common methods of microscopy. The glandular trichomes are composed of a secretory head with its cells in columnar format. The stalk is formed by two layers of cells, with the upper layer composed of cuboidal cells where the wall starts to thicken at the beginning of the pre-secretory phase. The secretion is heterogeneous, releasing glucose, other carbohydrates, lipids and phenolic compounds, with two types of release - eccrine and granulocrine. These trichomes are functionally termed as nectaries. The stalk cells appear as transfer cells since they have a thicker anticlinal wall with irregular ingrowths. The presence of transfer cells in the nectaries suggests a high specialization because it improves transport capacity of nectar and compensation in the high energy expenditure for its production and release.

Geographic variation in skull shape of the water rat Scapteromys tumidus (Cricetidae, Sigmodontinae): isolation-by-distance plus environmental and geographic barrier effects?
ABSTRACT The geographic variation in skull size and shape of the swamp rat Scapteromys tumidus was examined in samples from eight geographic clusters in almost of its distribution in southern Brazil and Uruguay. For analysis we used two-dimensional geometric morphometric methods for dorsal, ventral and lateral views of the skull. The geometric descriptors showed no significant differences in skull size between geographic clusters, while differences in shape were highly significant. We found a significant and moderate correlation between geographic and morphological distances, corroborating the isolation-by-distance model. Samples from the Rio Grande do Sul central coastal plain were the most differentiated, segregating completely from all other samples in canonical variate analysis for the dorsal view. The most visible variable regions in skull were the zygomatic arch (mainly the squamosal root of zygomatic) and the lateral braincase borders. Once correlation between geographic and morphological distances were not strong, it is possible that other factors (environmental heterogeneity and/or geographic barriers) may are acting in S. tumidus skull differentiation.

Objective measurement of Akodon budini dorsal coloration: methodological concerns
ABSTRACT The role of color in taxonomic and systematic studies of several taxa is central. Color data are more valuable if they permit comparisons. We aimed to provide pelage objective color measurements of a series of Akodon budini and to use those color data to test and to quantitatively analyze the influence of both the lighting source and the measuring point, in the determination of pelage color of museum skins. We used a spectroradiometer to measure the pelage color at five points over the dorsal midline of 54 Akodon budini museum skins. PCAs and ANOVAs were conducted over the color data. The characteristics of the studied series also allowed us to include and assess the potential effects of some main sources of intra-specific variation. Determinations of fur color strongly depend on the lighting source, and therefore it is essential that lighting conditions are controlled during color measurements and then made explicit when communicating the color characterization of particular taxonomic units. Furthermore, color determinations strongly depend on the measuring point, even on the same body area, and therefore it is critical that the color characterization of a taxon includes the variation between different points of the different body parts of the studied specimens.

Occurrence of annual growth rings in Rhizophora mangle in a region with low climate seasonality
ABSTRACT The formation of annual growth rings has been confirmed for several mangrove species in the last decade, among which is the Rhizophora mangle. However, the record of annual rings for this species was made in a region with high hydric seasonality, a widely recognized induction factor of annual rings in tropical species. In this sense, the present study aimed to verify the occurrence of annual growth rings in R. mangle in the mangroves of Guaratiba (Rio de Janeiro, Southeastern Brazil), a region with low hydric seasonality. For this purpose, the crossdating technique was applied in ten trees collected with known age (seven years). The growth rings are characterized by alternating layers of low vessel density (earlywood) and high vessel density (latewood). Multiple regression analysis indicated that growth rings width variation is driven by precipitation, water surplus, water deficit and water storage. Crossdating analysis confirmed the existence of annual growth rings in the R. mangle in Guaratiba. This discovery in a region with low hydric seasonality increases the dendrocronological potential of this species and suggests the importance of biological factors (eg. phenological behavior) as complementary inductors for the formation of growth rings in this species.

Chlorophyll fluorescence varies more across seasons than leaf water potential in drought-prone plants
ABSTRACT Among the effects of environmental change, the intensification of drought events is noteworthy, and tropical vegetation is predicted to be highly vulnerable to it. However, it is not clear how tropical plants in drought-prone habitats will respond to this change. In a coastal sandy plain environment, we evaluated the response of six plant species to water deficits across seasons, the relationship between their morpho-physiological traits, and which traits would be the best descriptors of plants' response to drought. Regardless of leaf succulence and phenology, responses between seasons were most strongly related to chlorophyll fluorescence. In this study we have demonstrated that a better comprehension of how tropical species from drought-prone habitats cope with changes in water availability can be based on seasonal variation in leaf water potential and chlorophyll fluorescence. Temporal variation in leaf water potential and chlorophyll fluorescence was found useful for differentiating between groups of sandy soil species that are responsive or unresponsive to water availability. However, chlorophyll fluorescence appeared to be a more sensitive descriptor of their seasonal and short-term responses.

Effect of osmopriming on germination and initial growth of Physalis angulata L. under salt stress and on expression of associated genes
ABSTRACT This study aimed to evaluate the effects of priming on seed germination under salt stress and gene expression in seeds and seedlings of P. angulata L. After priming for 10 days, seed germination was tested in plastic trays containing 15 ml of water (0 dS m-1 - control) or 15 ml of NaCl solution (2, 4, 6, 8, 10, 12, 14 and 16 dS m-1). Fresh and dry weight of shoots and roots of seedlings were evaluated at 0, 2, 4, 6, 8 dS m-1. Total RNA was extracted from whole seeds and seedlings followed by RT-qPCR. The target genes selected for this study were: ascorbate peroxidase (APX), glutathione-S-transferase (GST), thioredoxin (TXN), high affinity potassium transporter protein 1 (HAK1) and salt overly sensitive 1 (SOS1). At an electroconductivity of 14 dS m-1 the primed seeds still germinated to 72%, in contrast with the non-primed seeds which did not germinate. The relative expression of APX was higher in primed seeds and this may have contributed to the maintenance of high germination in primed seeds at high salt concentrations. GST and TXN displayed increased transcript levels in shoots and roots of seedlings from primed seeds. Priming improved seed germination as well as salt tolerance and this is correlated with increased expression of APX in seeds and SOS1, GST and TXN in seedlings.

Production, purification and characterization of an exo-polygalacturonase from Penicillium janthinellum sw09
ABSTRACT A soil isolate, Penicillium janthinellum sw09 has been found to produce significant amounts of an extracellular pectinase subsequently characterized as exo-polygalacturonase (exo-PG). By optimizing growth conditions, P. janthinellum sw09 produced high amount of exo-PG (16.54 units/mL). The crude enzyme was purified by gel filtration chromatography and two exo-PG activity peaks (designated as PGI and PGII) were revealed. On SDS-PAGE analysis, purified PGII using DEAE-Sepharose FF column, was found to be a single band with a molecular mass of 66.2 kDa. The purified PGII exhibited maximal activity at the temperature of 45 oC and pH 5.0. The stability profiles show that PGII is more stable in the pH range of 4.0-8.0 and below 60 oC. The Km and Vmax for the enzyme was 1.74 mg/mL and 18.08 μmol/ (mL•min), respectively. Due to this enzymatic characterization, this pectinase is an attractive candidate for applications in degradation of pectin.

The effect of exotic grass Urochloa decumbens (Stapf) R.D.Webster (Poaceae) in the reduction of species richness and change of floristic composition of natural regeneration in the Floresta Nacional de Carajás, Brazil
ABSTRACT The introduction of exotic species is considered as one of the major causes of biodiversity loss. The National Forest of Carajás is one of the largest mineral provinces in the world. Mining activities caused changes of the natural habitats, leaving degraded areas after the mineral exploitation. One of the mining areas within FLONA Carajás was used for the extraction of gold. In the process of exploitation, a huge depression was formed by the removal of soil which was mounded up nearby. To prevent soil erosion of these mounds, an exotic grass, Urochloa decumbens (Stapf) R.D.Webster (Poaceae) was planted. The objective of this study was to compare the impact of this non-native grass on species richness and species composition of the natural regeneration in the degraded areas. Four areas were compared, two with and two without presence of U. decumbens. In each area, twenty four 1m²/plots were established. Species richness of the regeneration areas and population sizes were significantly lower in the plots where the exotic grass was present. Our study shows that U. decumbens had a negative effect on species richness and population density, and its presence changed the species composition and distribution of life forms of the natural regeneration.

Detection and quantification of human adenovirus genomes in Acanthamoeba isolated from swimming pools
ABSTRACT Acanthamoeba is the most common free-living environmental amoeba, it may serve as an important vehicle for various microorganisms living in the same environment, such as viruses, being pathogenic to humans. This study aimed to detect and quantify human adenoviruses (HAdV) in Acanthamoebas isolated from water samples collected from swimming pools in the city of Porto Alegre, Southern Brazil. Free-living amoebae of the genus Acanthamoeba were isolated from water samples, and isolates (n=16) were used to investigate the occurrence of HAdVs. HAdV detection was performed by quantitative real-time polymerase chain reaction (qPCR). HAdVs were detected in 62.5% (10/16) of Acanthamoeba isolates, ranging from 3.24x103 to 5.14x105 DNA copies per milliliter of isolate. HAdV viral loads found in this study are not negligible, especially because HAdV infections are associated with several human diseases, including gastroenteritis, respiratory distress, and ocular diseases. These findings reinforce the concept that Acanthamoeba may act as a reservoir and promote HAdV transmission through water.

Molecular biology of mycoplasmas: from the minimum cell concept to the artificial cell
ABSTRACT Mycoplasmas are a large group of bacteria, sorted into different genera in the Mollicutes class, whose main characteristic in common, besides the small genome, is the absence of cell wall. They are considered cellular and molecular biology study models. We present an updated review of the molecular biology of these model microorganisms and the development of replicative vectors for the transformation of mycoplasmas. Synthetic biology studies inspired by these pioneering works became possible and won the attention of the mainstream media. For the first time, an artificial genome was synthesized (a minimal genome produced from consensus sequences obtained from mycoplasmas). For the first time, a functional artificial cell has been constructed by introducing a genome completely synthesized within a cell envelope of a mycoplasma obtained by transformation techniques. Therefore, this article offers an updated insight to the state of the art of these peculiar organisms' molecular biology.

Molecular and cytogenetic assessment of Dipterygium glaucum genotoxicity
ABSTRACT The aim of the present study is to assess the genotoxicity of Dipterygium glaucum grows widely in Saudi Arabia desert to produce safety herbal products. This work is considered the first and pioneer report so far due to the lack and poor evaluated reports of the plant species for their mutagensity, genotoxicity and cytogenetics effects. Cytogenetic effects of D. glaucum on mitotic in roots of Vicia faba showed reduction in mitotic activity using three extracts; water, ethanol and ethyl acetate. Chromosomal abnormalities were recorded that included stickiness of chromosomes, chromatin bridge, fragments, lagging chromosome and micronuclei. Protein bands and RAPD analyses of V. faba treated with three D. glaucum extracts revealed some newly induced proteins and DNA fragments and other disappeared. Chemical constitution of the plant species should be identified with their biological activities against human and animal cells like HeLa cancer cell line. We are recommending using additional genotoxicity tests and other toxicity tests on animal culture with different concentrations and also utilizing several drought and heat tolerant genes of the plant species in gene cloning to develop and improve other economical crop plants instead of using the species as oral herbal remedy

Differential effects in CGRPergic, nitrergic, and VIPergic myenteric innervation in diabetic rats supplemented with 2% L-glutamine
ABSTRACT The objective of this study was to investigate the effects of 2% L-glutamine supplementation on myenteric innervation in the ileum of diabetic rats, grouped as follows: normoglycemic (N); normoglycemic supplemented with L-glutamine (NG); diabetic (D); and diabetic supplemented with L-glutamine (DG). The ileums were subjected to immunohistochemical techniques to localize neurons immunoreactive to HuC/D protein (HuC/D-IR) and neuronal nitric oxide synthase enzyme (nNOS-IR) and to analyze varicosities immunoreactive to vasoactive intestinal polypeptide (VIP-IR) and calcitonin gene-related peptide (CGRP-IR). L-Glutamine in the DG group (i) prevented the increase in the cell body area of nNOS-IR neurons, (ii) prevented the increase in the area of VIP-IR varicosities, (iii) did not prevent the loss of HuC/D-IR and nNOS-IR neurons per ganglion, and (iv) reduced the size of CGRP-IR varicosities. L-Glutamine in the NG group reduced (i) the number of HuC/D-IR and nNOS-IR neurons per ganglion, (ii) the cell body area of nNOS-IR neurons, and (iii) the size of VIP-IR and CGRP-IR varicosities. 2% L-glutamine supplementation exerted differential neuroprotective effects in experimental diabetes neuropathy that depended on the type of neurotransmitter analyzed. However, the effects of this dose of L-glutamine on normoglycemic animals suggests there are additional actions of this beyond its antioxidant capacity.

Effects of Tityus stigmurus (Thorell 1876) (Scorpiones: Buthidae) venom in isolated perfused rat kidneys
ABSTRACT Scorpions belonging to the Tityus genus are of medical interest in Brazil. Among them, Tityus stigmurus is the main scorpion responsible for stings in the Northeast region. After a sting, the scorpion venom distributes rapidly to the organs, reaching the kidneys quickly. However, there are few studies concerning the renal pathophysiology of scorpion poisoning. In this study, we evaluated the effects of T. stigmurus venom (TsV) on renal parameters in isolated rat kidneys. Wistar rats (n = 6), weighing 250-300 g, were perfused with Krebs-Henseleit solution containing 6 g/100 mL bovine serum albumin. TsV at 0.3 and 1.0 μg/mL was tested, and the effects on perfusion pressure (PP), renal vascular resistance (RVR), urinary flow (UF), glomerular filtration rate (GFR), and electrolyte excretion were analyzed. Effects were observed only at TsV concentration of 1.0 μg/mL, which increased PP (controlPP40' = 92.7 ± 1.95; TsVPP40' = 182.0 ± 4.70* mmHg, *p < 0.05), RVR (controlRVR40' = 3.28 ± 0.23 mmHg; TstRVR40' = 6.76 ± 0.45* mmHg, *p < 0.05), UF (controlUF50' = 0.16 ± 0.04; TstUF50' = 0.60 ± 0.10* mL/g/min,*p < 0.05), GFR and electrolyte excretion, with histological changes that indicate renal tubular injury. In conclusion, T. stigmurus venom induces a transient increase in PP with tubular injury, both of which lead to an augmented electrolyte excretion.

Lizard assemblage from a sand dune habitat from southeastern Brazil: a niche overlap analysis
ABSTRACT Communities are structured by interactions of historical and ecological factors, which influence the use of different resources in time and space. We acquired data on time of activity, microhabitat use and diet of a lizard assemblage from a sand dune habitat in a coastal area, southeastern Brazil (Restinga de Jurubatiba). We analyzed the data of niche overlap among species in these three axes (temporal, spatial and trophic) using null models. We found a significant overlap within the trophic niche, whereas the overlap for the other axes did not differ from the expected. Based on this result, we discuss the factors acting on the structure of the local lizard community.

Flood regime and water table determines tree distribution in a forest-savanna gradient in the Brazilian Pantanal
ABSTRACT This study aimed to recognized the preferential location of species of the tree sinusiae in response to a moisture gradient in Pantanal Matogrossense, Brazil. We established sampling plots of arboreal sinusiae along a soil moisture and flood gradient. Piezometers were installed, allowing monthly measurements of water table depth and flood height during one year. Detrended Correspondence Analysis, Gradient Direct Analysis, Multi-response Permutation Procedures and Indicator Species Analysis were performed to evaluate the effect of moisture gradient on tree distribution. The annual variation of water table is shallower and similar in Seasonally Flooded Forest and Termite Savanna, with increasing depths in Open Savanna, Savanna Forest and Dry Forest. Circa 64% of the species were characterized as having a preferential location in "terrestrial habitats normally not subjected to inundation", while 8% preferentially occur in "wet habitats". Lowest tree richness in flood-affected vegetation types is related to both present-day high climatic seasonality and Late Pleistocene dry paleoclimates in the Pantanal wetland. The tree distribution across different formations in the Pantanal shows a direct relationship with soil moisture gradient.

A phytosociological analysis and synopsis of the dry woodlands and succulent vegetation of the Peruvian Andes
ABSTRACT A phytosociological approach to dry forest and cactus communities on the occidental slopes of the Peruvian Andes is presented in base of 164 plots carried out following the Braun-Blanquet method. From them, 52 have been made recently, and the other 112 were taken from the literature. After a multivariate analysis, using a hierarchical clustering and a detendred correspondence analysis, the Acacio-Prosopidetea class (dry forest and cactus communities, developed on soils with some edaphic humidity or precipitations derived from El Niño Current), the Opuntietea sphaericae class (cactus communities of central and southern Peru, on few stabilized rocky or sandy soils) and the Carico-Caesalpinietea class (dry forests of the Peruvian coastal desert, influenced by the maritime humidity of the cold Humboldt Current), are differentiated. Within the Acacio-Prosopidetea class, two alliances are commented: the Bursero-Prosopidion pallidae (with two new associations Loxopterygio huasanginis-Neoraimondietum arequipensis and Crotono ruiziani-Acacietum macracanthae), and the new alliance Baccharido-Jacarandion acutifoliae (with the new associations Armatocereo balsasensis-Cercidietum praecocis and Diplopterydo leiocarpae-Acacietum macracanthae). For the Opuntietea sphaericae class, the association Haageocereo versicoloris-Armatocereetum proceri (Espostoo-Neoraimondion) is described on the basis of plots from hyperarid localities of central Peru. Finally, a typological classification of the studied plant communities is given.

Diversity and associations between Drosophilidae (Diptera) species and Basidiomycetes in a Neotropical forest
ABSTRACT Drosophilidae is one of the most representative families of insects that occurs in fungal fruiting bodies of Basidiomycetes; however, the diversity and community structure of mycophagous Drosophilidae in the Neotropical region is poorly known. The aims of the present study were to describe the diversity of mycophagous Drosophilidae and to investigate its colonization of fungal hosts in a forest of southern Brazil. From 120 fungal samples (patches of mushrooms) of 17 Basidiomycetes genera, flies were recorded emerging from 70 samples and collected in adult stages of 25 fungal samples, for a total of 4897 drosophilids belonging to 31 species and 5 genera. Drosophila Fallén was the most species-rich genus, whereas Hirtodrosophila Duda was the dominant genus. Studies performed in the Holarctic region indicate that mycophagous drosophilid have generalist habits; however, our results showed that most drosophilids use fewer than two fungal hosts, and most species of Hirtodrosophila and Leucophenga were restricted to abundant fungal species, suggesting a specialization for these resources. The most specialized fauna emerged from Auricularia, which was the most frequent fungal genus in our collection, and this result supports the assumption that specialization depends on the availability of fungal resources over time.

Ultrastructure and pollen morphology of Bromeliaceae species from the Atlantic Rainforest in Southeastern Brazil
Pollen grain morphology of Bromeliaceae species collected in areas of the Atlantic Rainforest of southeastern Brazil was studied. The following species were analyzed: Aechmea bambusoides L.B.Sm. & Reitz, A. nudicaulis (L.) Griseb., A. ramosa Mart. ex Schult.f., Ananas bracteatus (Lindl.) Schult.f., Billbergia distachia (Vell.) Mez, B. euphemiae E. Morren, B. horrida Regel, B. zebrina (Herb.) Lindl., Portea petropolitana (Wawra) Mez, Pitcairnia flammea Lindl., Quesnelia indecora Mez, Tillandsia polystachia (L.) L., T. stricta Sol., T. gardneri Lindl., T. geminiflora Brongn. and Vriesea grandiflora Leme. Light and scanning electron microscopy were used and the species were grouped into three pollen types, organized according to aperture characteristics: Type I - pantoporate pollen grains observed in P. petropolitana, Type II - 2-porate pollen grains, observed in the genera Ananas, Aechmea and Quesnelia, and Type III - 1-colpate pollen grains, observed in the genera Billbergia, Pitcairnia, Tillandsia and Vriesea. Pollen data led to the construction of an identification key. The results showed that the species analyzed can be distinguished using mainly aperture features and exine ornamentation, and that these characteristics may assist in taxonomic studies of the family.

Life cycle traits of Philodina roseola Ehrenberg, 1830 (Rotifera, Bdelloidea), a model organism for bioassays
This paper describes experimental results on the life cycle of the rotifer Philodina roseola cultured in the laboratory. Detailed information on life-cycle parameters of a certain species provides a deep understanding and contributes to a better knowledge of the role of the species in the community, besides providing data that are basic to other ecological investigations such as secondary production estimates and knowledge for applications such as its utilization as test-organism in ecotoxicological studies. The average duration of embryonic development of P. roseola was 23.88 h, the age at maturity of primipara was 3.5 days and the maximum lifespan was 23 days. The average size of the rotifer neonate was 198.77 µm, the mean size of primipara was 395.56 µm and for adults 429.96 µm. The average fecundity was 1.22 eggs per female per day and the mean number of eggs produced per female during the entire life was 22.33. The deceleration of somatic growth from the start of the reproductive stage represents a trade-off between growth and reproduction that is often seen in micrometazoans. The life history of P. roseola follows the strategy of other bdelloid species characterized by a rapid pre-reproductive development and canalization of most assimilated energy to reproduction after reaching maturity. The differences observed in total fecundity and longevity between our P. roseola cultures and those from previous studies were probably due to differences of intrinsic adaptation of this species ecotypes to the conditions of their natural environments.

Hippocampal distribution of IL-1β and IL-1RI following lithium-pilocarpine-induced status epilepticus in the developing rat
The contribution of Interleukin-1β (IL-1β) to neuronal injury induced by status epilepticus (SE) in the immature brain remains unclear. The goal of this study was to determine the hippocampal expression of IL-1β and its type 1 receptor (IL-1RI) following SE induced by the lithium-pilocarpine model in fourteen-days-old rat pups; control animals were given an equal volume of saline instead of the convulsant. IL-1β and IL-1RI mRNA hippocampal levels were assessed by qRT-PCR 6 and 24 h after SE or control conditions. IL-1β and IL-1RI expression was detected in the dorsal hippocampus by immunohistochemical procedures; Fluoro-Jade B staining was carried out in parallel sections in order to detect neuronal cell death. IL-1β mRNA expression was increased 6 h following SE, but not at 24 h; however IL-1RI mRNA expression was unaffected when comparing with the control group. IL-1β and IL-1RI immunoreactivity was not detected in control animals. IL-1β and IL-1RI were expressed in the CA1 pyramidal layer, the dentate gyrus granular layer and the hilus 6 h after SE, whereas injured cells were detected 24 h following seizures. Early expression of IL-1β and IL-1RI in the hippocampus could be associated with SE-induced neuronal cell death mechanisms in the developing rat.
