Reproductive phenology in a riparian rainforest in the south of Santa Catarina state, Brazil

Phenological studies assist in forest ecosystems comprehension and evaluation of resource availability for wildlife, as well as in improving the understanding of relationships between plants and their pollinators and dispersers. This study aims to describe the reproductive phenophases of riparian plant species and correlate them with climatic variables. The reproductive phenology was analyzed biweekly throughout one year, recording the absence or presence of flowers/fruits. The flowering phenophase occurred throughout the year, with an increase in number of species in blossom in October, November, and December. The flowering peak of the community was observed in November. The fruiting phenophase also occurred throughout the year and showed an increase of species fruiting in June with a slight decrease in August and September. The data obtained in this study, when compared with other studies in different Atlantic Rainforest areas, indicates a seasonal pattern for the flowering phenophase and a variation in fruit availability throughout the year as well as in the fruiting peaks. Therefore, studies that observe flowering and fruiting events in loco are of main importance because they provide information on reproductive seasons of species for use in environmental restoration projects and thus alleviate the situation of degradation of riparian forests.

Is the secondary thickening in palms always diffuse?

Unlike other arboreal monocotyledons, the secondary growth of palms has for the past 100 years been described as diffuse. Solely cell enlargement and random parenchyma divisions, without the activity of a meristem, characterize such growth. Some previous works of the early 20th century have, however, mentioned the presence of a secondary meristem in the stems of palms, but this information was forgotten since then. Addressing to this question, we analysed palm stems of four species, with the aim to understand the possible presence of such secondary growth. We found that a meristematic band occurs between the cortex and the central cylinder and gives rise to new vascular bundles and parenchyma internally, producing parenchyma and fibres externally. It appears secondarily, i.e., it undergoes meristematic activity in the median and basal stem regions, far away from the apical region. In fact, a meristematic band is present and may be more common than currently believed, but uneasy to detect in certain palms for being restricted to specific regions of their stems. In conclusion, the diffuse secondary thickening is here shown not to be the only mechanism of secondary growth in palms. The presence of a meristem band in the stems of palms merits careful reconsideration.

Fermentation characteristics in stirred-tank reactor of exopolysaccharides with hypolipidemic activity produced by Pleurotus geesteranus5#

In this study, the hypolipidemic effect of exopolysaccharides (EPS) fromPleurotus geesteranus 5# fermenting liquor by the optimal culture conditions in a 5-L stirred-tank reactor was investigated. The hypolipidemic effect of the polysaccharide, investigated in streptozotocin induced diabetic mice, decreased plasma glucose, total cholesterol and triacylglycerol concentrations by 17.1 %, 18.8 % and 12.0 %, respectively. The results of the present investigation strongly demonstrate the potential of this polysaccharide to prevent hyperglycemia in the experimental animals. Under optimal culture conditions, the maximum concentrations of mycelial and EPS were 22.63 g/L after 7 d cultivation and 11.09 g/L after 10 d, respectively. Furthermore, the morphological parameters (i.e. mean diameter, circularity, roughness and compactness) of the pellets and the broth viscosity were characterized. It was proved that compactness of the pellet morphology (R2=0.963, p<0.01) was significantly and positively determined with mycelial biomass. Moreover, mean diameter (R2=93.3, p<0.01) and broth viscosity (R2=0.950, p<0.01) were significantly and positively determined with EPS content.

Standardization of a method to study angiogenesis in a mouse model

In the adult organism, angiogenesis is restricted to a few physiological conditions. On the other hand, uncontrolled angiogenesis have often been associated to angiogenesis-dependent pathologies. A variety of animal models have been described to provide more quantitative analysis ofin vivo angiogenesis and to characterize pro- and antiangiogenic molecules. However, it is still necessary to establish a quantitative, reproducible and specific method for studies of angiogenesis factors and inhibitors. This work aimed to standardize a method for the study of angiogenesis and to investigate the effects of thalidomide on angiogenesis. Sponges of 0.5 x 0.5 x 0.5 cm were implanted in the back of mice groups, control and experimental (thalidomide 200 mg/K/day by gavage). After seven days, the sponges were removed. The dosage of hemoglobin in sponge and in circulation was performed and the ratio between the values was tested using nonparametric Mann-Whitney test. Results have shown that sponge-induced angiogenesis quantitated by ratio between hemoglobin content in serum and in sponge is a helpful model for in vivo studies on angiogenesis. Moreover, it was observed that sponge-induced angiogenesis can be suppressed by thalidomide, corroborating to the validity of the standardized method.

Tau, APP, NCT and BACE1 in lymphocytes through cognitively normal ageing and neuropathology

Although Alzheimer's disease is a brain disorder, a number of peripheral alterations have been found in these patients; however, little is known about how the key genes involved in the pathophysiology express in peripheral cells such as lymphocytes during normal compared to neuropathological ageing. We analysed the expression of tau, of the amyloid precursor protein, of nicastrin and of the β-site APP cleaving enzyme genes by RT-PCR in lymphocytes from a small group of late-onset Alzheimer's disease patients, from aged patients suffering from neuropsychological conditions different from Alzheimer's and from cognitively healthy subjects divided in four groups by age. We also investigated correlations between gene expression and levels of blood pressure, glucose, total cholesterol and triglycerides as risk factors for Alzheimer's. Results show no tau expression in lymphocytes, a lack of detection of nicastrin expression in Alzheimer's patients and correlations between the medical conditions studied and gene expression in lymphocytes. We believe nicastrin gene expression in lymphocytes should be considered of interest for further analyses in a wider population to investigate whether it might represent a potential biomarker to differentiate Alzheimer's from other neuropsychological disorders.

Evaluation of Bcl-2, Bcl-x and Cleaved Caspase-3 in Malignant Peripheral Nerve Sheath Tumors and Neurofibromas

ABSTRACTAIMSTo study the expression of Bcl-2, Bcl-x, as well the presence of cleaved caspase-3 in neurofibromas and malignant peripheral nerve sheath tumors. The expression of Bcl-2 and Bcl-x and the presence of cleaved caspase 3 were compared to clinicopathological features of malignant peripheral nerve sheath tumors and their impact on survival rates were also investigated.MATERIALS AND METHODSThe evaluation of Bcl-2, Bcl-x and cleaved caspase-3 was performed by immunohistochemistry using tissue microarrays in 28 malignant peripheral nerve sheath tumors and 38 neurofibromas. Immunoquantification was performed by computerized digital image analysis.CONCLUSIONSApoptosis is altered in neurofibromas and mainly in malignant peripheral nerve sheath tumors. High levels of cleaved caspase-3 are more common in tumors with more aggressive histological features and it is associated with lower disease free survival of patients with malignant peripheral nerve sheath tumors.

Nutritional parameters of steers receiving different levels of sunflower crushed in partial replacement of soybean meal

ABSTRACTTo evaluate of the sunflower crushed in nutritional parameters in steers, supplemented at pasture, we used four steers in 4×4 Latin square design. The supplements were provided in 6 g-kg of body weight-animal-day, consisting of sunflower crushed, corn, soybean meal and mineral. All the supplements was isonitrogenous and soybean meal was replaced in 0, 20, 40, and 60% for sunflower crushed. The determination of ruminal pH and ammonia was at 0, 2, 4, 6 and 8 h. after feeding and for short-chain fatty acids it was collected at 0 and 6 h. post-feeding. The dry matter intake was not affected (P>0.05) by inclusion of sunflower crushed (mean=6.59 kg-day). There was no significant effect (P>0.05) for pH for the inclusion of sunflower crushed (mean=6.41). For contents of ruminal NH3-N was a significant effect (P <0.05) only for collection time, and ammonia peaks occurred between 2 and 4 h after feeding, with values of 22.56 and 21.40 mg-dL. The total concentration of short chain fatty acids and the C2:C3 ratio was reduced in 9.6 and 15.43%. The ruminal degradability of NDF was not affected by the supplements. The supplementation with sunflower crushed to beef steers grazing, in partial replacement of soybean meal did not alter nutrition parameters.

Assessing rainfall erosivity indices through synthetic precipitation series and artificial neural networks

The rainfall parameter that expresses the capacity to promote soil erosion is called rainfall erosivity (R), and is commonly represented by the indexes EI30 and KE>25. The calculations of these indexes requires pluviographical records, that are difficult to obtain in Brazil. This paper describes the use of synthetic rainfall series to compute EI30 and KE>25 in Espírito Santo State (Brazil). Artificial neural networks (ANNs) were also developed to spatially interpolate R values in Espírito Santo. EI30 and KE>25 indexes values were close to those calculated on a homogeneous area according to the similarity of rainfall distribution; indicating the applicability of the use of synthetic rainfall series to estimate the R factor. ANNs had a better performance than Inverse Distance Weighted and Kriging to spatially interpolate rainfall erosivity values in the State of Espírito Santo.

Centesimal composition and physical-chemistry analysis of the edible mushroom Lentinus strigosus occurring in the Brazilian Amazon

The centesimal composition and the physical and chemical analyses of Lentinus strigosus, an edible mushroom occurring in the Brazilian Amazon and produced in alternative substrates based on wood and agroindustrial residues, were evaluated. For this purpose, the C, N, pH, soluble solids, water activity, protein, lipids, total fiber, ash, carbohydrate, and energy levels were determined. The substrates were formulated from Simarouba amara Aubl. (“marupá”), Ochroma piramidale Cav. Ex. Lam. (“pau-de-balsa”) and Anacardium giganteum (“cajuí”) sawdust and Bactris gasipaes Kunth (“pupunheira”) stipe and Saccharum officinarum (sugar cane bagasse). The results indicated that the nutritional composition of L. strigosus varied with the substrate of cultivation; the protein levels found in mushrooms grown in the different substrates (18 – 21.5%) varied with the substrate and was considered high; the soluble solids present in the mushrooms could have a relation with complex B hydrosoluble vitamins. L. strigosus could be considered as important food owing to its nutritional characteristics such as high protein content, metabolizable carbohydrates and fibers, and low lipids and calories content.

Induction and characterization of oil palm (Elaeis guineensis Jacq.) pro-embryogenic masses

ABSTRACTOil palm is one of the most economically valuable oil seed plants, but the expansion of plantations has been limited by availability of seedlings, as the conventional propagation is through seeds, which have low germination rates. One possible solution for the large-scale production is the use of somatic embryogenesis. The aim of this study was evaluate the effects auxins 2,4-D and picloram on the induction of pro-embryogenic masses inE.guineenesis hybrid leaf explants and characterize, regarding embryogenic characteristics, with cytochemical and ultrastructural analisys. Specifically, in vitro plantlets leaves fragments were inoculated in Y3 culture medium supplemented by 2.4-D or picloram at different concentrations (0.0, 1.0, 3.0, 6.0 and 9.0 mg l–1). After 90 days the presence- absence of cell masses were evaluated. Both growth regulators efficiently induced cellular masses regardless of the concentrations applied. As the cell masses were not homogeneously formed, they were classified according to color and shape into four types: TYPE 1 - elongated and translucent, TYPE 2 – uneven and translucent, TYPE 3 - globular and beige TYPE 4 – globular and white. Based on the anatomical and ultrastructural features, TYPE 2, 3 and 4 cell masses were considered to have the highest embryogenic potential and therefore may be most suited to large-scale vegetative propagation of oil palm.

Preliminary phytochemical screening and molluscicidal activity of the aqueous extract of Bidens pilosa Linné (Asteraceae) in Subulina octona (Mollusca, Subulinidade)

The aim of this study was to establish reference concentrations (LC50 e LC90) of aqueous extracts of Bidens pilosa on the land snail Subulina octona, in order to assess the changes caused by 24 and 48 h exposure to the sublethal concentration regarding species' fecundity, hatchability and in offspring produced after exposure to confirm the presence of tannins, saponins and flavonoids in this plant species. Eggs newly hatched and 30 day-old snails were exposed to sublethal concentration, calculated for adults. The phytochemical analysis confirmed the presence of flavonoids, condensed tannins and saponins in the aqueous extract of B. pilosa. The LC50 and LC90 obtained were 51.4mg-mL and 74.1mg-mL respectively. The exposure to sublethal concentration reduced significantly the hatchability and the survival of the offspring hatched from exposed eggs and also caused a reduction on survival and growth in snails exposed for both time period. The exposure time also caused a reduction at the evaluated parameters.

Spatial and temporal analysis of stem bleeding disease in coconut palm in the state of sergipe, Brazil

ABSTRACTStem bleeding disease (resinosis) of coconut palm is caused by Thielaviopsis paradoxa and is very important in the state of Sergipe, Brazil. Understanding the epidemiological behavior of the disease is essential for establishing more efficient control strategies. Thus, we characterized the temporal progression and spatial distribution of stem bleeding in a commercial orchard under conditions of natural infection in the area of Neopolis, Sergipe. Three plots with 729 plants each were selected and evaluated every two months for stem bleeding incidence. In the temporal analysis, the monomolecular model gave the best fit to data on disease incidence, as it accurately showed the temporal dynamics of the disease during the experiment period. The spatial pattern of stem bleeding varied over time, with initial infections presenting random pattern and then evolving to aggregate pattern during evaluations. This indicates that the disease may have originated from the pathogen survival structures, followed by auto infections caused by dissemination from plant to plant, either by humans, by contact between roots, or by the vector Rhynchophorus palmarum.

Perception of local inhabitants regarding the socioeconomic impact of tourism focused on provisioning wild dolphins in Novo Airão, Central Amazon, Brazil

Botos (Inia geoffrensis) are currently provisioned for use in tourist attractions in five sites in the Brazilian Amazon. Despite the known negative effects associated with human-wild dolphin interactions, this activity has been regulated and licensed in the Anavilhanas National Park in Novo Airão, Amazonas State, Brazil. We present an updated evaluation of the perception of the local community concerning the possible socioeconomic impacts of this tourism in Novo Airão. In April 2011, 45 interviews were conducted with inhabitants. A small segment of Novo Airão perceives currently itself as being economically dependent on the botos feeding tourism. Despite that, the economic benefits of this controversial activity apparently are not shared among most inhabitants, and botos feeding tourism is perceived as generating diverse negative effects. We conclude that if the activity was banned or modified into a less impacting tourist activity, this action would probably not majorly affect the lives of the general population.

E-commerce adoption in Malaysian Small and Medium Enterprises Practitioner Firms: A revisit on Rogers' model

The current climate of business necessitates competitions that are often tough and unpredictable. All organizations, regardless their size and scope of operation, are facing severe competitive challenges. In order to cope with this phenomenon, managers are turning to e-commerce in their respective organizations. The present study hinges upon exploring and explaining the different dimensions of the adoption of e-commerce among Small and medium enterprises, based on the Five Factors of Diffusion of Innovation Model derived by Rogers. In this study, however, we employed the survey methods. A questionnaire was distributed to 1,200 managers and employees in the manufacturing, service and agricultural sectors by email; with a response rate of 10%. The results gleamed from this study posits that relative advantage is influential vis-à-vis e-commerce adoption. Trialability and Observability factors affect the level of confidence of management, which in turn, influences e-commerce adoption. Meanwhile, the existing culture of a company affects the resistance of employees, which in turn negatively effects the e-commerce adoption, while complexity does not significantly influence the e-commerce adoption.

Epigallocatechin-3-gallate combined with alpha lipoic acid attenuates high glucose-induced receptor for advanced glycation end products (RAGE) expression in human embryonic kidney cells

The anti-oxidant effects of epigallocatechin gallate (EGCG) and alpha lipoic acid (ALA) have been demonstrated in previous studies. The kidney protection effects of EGCG and ALA in patients with kidney injury are still under investigation. The purpose of this study is to investigate the anti-inflammatory and anti-oxidant effects of EGCG and ALA on high glucose-induced human kidney cell damage. EGCG inhibited high glucose(HG)-induced TNF-α and IL-6 production in human embryonic kidney (HEK) cells. Both EGCG and ALA decreased HG-induced receptor of advanced glycation end products (RAGE) mRNA and protein expressions in HEK cells. EGCG and ALA also recovered HG-inhibited superoxide dismutase production and decreased ROS expressions in HEK cells. The synergism of EGCG and ALA was also studied. The effect of EGCG combined with ALA is greater than the effect of EGCG alone in all anti-inflammation and anti-oxidant experiments. Our studies provide a potential therapeutic application of EGCG and ALA in preventing progression of diabetic nephropathy.

Production of 5-hydroxy-7-methoxy-4-methylphthalide in a culture of Penicillium crustosum

The chemical reactions carried out by microorganisms have been used as a tool in modern chemistry. This paper reports the production of mycophenolic acid and a new phthalide by the endophytic fungus Penicillium crustosum obtained from coffee seeds. The fungus was cultivated in a liquid medium for a period of seven days and after that the culture medium was divided into four treatments: A, B, C and D, to which different organic substances were added. Treatment A was maintained as the control to evaluate the occurrence of biotransformation. Organic acids were added to the culture media of treatments B (ferulic and quinic acids) and C [cinnamic and 3,4-(methylenedioxy) cinnamic acids], and caffeine was added in the treatment D. All these organic compounds were dissolved in DMSO, and the fermentation was maintained for more 13 days, totalizing 20 days. Mycophenolic acid was isolated from the culture with no added acids (treatment A). Mycophenolic acid and a new phthalide, 5-hydroxy-7-methoxy-4-methylphthalide were isolated from treatments B and C, and mycophenolic acid and caffeine (added to the culture medium) were isolated from treatment D. The structures were determined by NMR techniques and confirmed by MS and MS/MS techniques.

Agro-economic yield of taro clones in Brazil, propagated with different types of cuttings, in three crop seasons

The experimental studies were conducted in 2007-2008, 2008-2009 and 2009-2010 crop seasons, in order to know the agro-economic yield of 'Chinês' and "Macaquinho" taro clones, propagated using huge, extra, large, medium, small and tiny cormels. The harvest was done on average on 202 days after planting, in three crop seasons. Based on the joint analysis of variance carried out, it was observed that taro clones showed significant differences in the yield of fresh and dry weight of leaves, cormels, and commercial and non-commercial comels; besides, there were significant differences in yield of a crop season to another and the size of the cuttings induced significant differences in yield. In the conditions that the experiments were conducted, and considering the highest average yield of fresh weight of commercial cormels (28.69 t.ha-1) and highest net income (US $14,741.14) correspondent to the three crop seasons, it is recommended to cultivate 'Macaquinho' clone using small cuttings in propagation.

Testing arbitrary classes of light in a physiognomically heterogeneous area of "campo rupestre" vegetation

The Brazilian "campo rupestre" vegetation shows high light heterogeneity, characteristic which is often disregarded in ecophysiological studies due the difficult of obtaining accurate measurements in a complex environment. The aim of the present study it was to describe how much heterogeneous is the light distribution in this environment, as well as tried to define classes in relation to the photosynthetically active radiation (PAR) for 30 rupicolous individuals of Bulbophyllum involutum. The individuals were marked and previously classified in light classes visually predetermined ("full sun", "partially shaded" and "shaded"). Once defined these classes, principal component analysis, cluster and dispersal analyses using direct and indirect PAR measures were performed to characterize the environment as well as to validate the inclusion of the individuals in each class. The use of multivariate analysis increased the accuracy of predetermined classes of light, and some individuals from "shaded" and "partially shaded" classes needed to be regrouped. After individuals' reclassification, amplitude of PAR variance was reduced in each class, enhancing homogeneity. Ours results show the possibility to evaluate light environment conditions more precisely in "campo rupestre" vegetation using multivariate analysis, allowing the increase in accuracy of this parameter for ecophysiological researches.

Hormesis with glyphosate depends on coffee growth stage

Weed management systems in almost all Brazilian coffee plantations allow herbicide spray to drift on crop plants. In order to evaluate if there is any effect of the most commonly used herbicide in coffee production, glyphosate, on coffee plants, a range of glyphosate doses were applied directly on coffee plants at two distinct plant growth stages. Although growth of both young and old plants was reduced at higher glyphosate doses, low doses caused no effects on growth characteristics of young plants and stimulated growth of older plants. Therefore, hormesis with glyphosate is dependent on coffee plant growth stage at the time of herbicide application.

Mathematic model of three-phase induction machine connected to advanced inverter for traction system for electric trolley

This paper establishes a mathematical model of induction machine connected to a frequency inverter necessary to adjust the electric motor drive. The mathematical model based on the Park's theory allows the analysis of the whole spectrum (electric car – frequency inverter) to drive the electric trolley bus made on ASTRA Bus Arad (Romania). To remove higher order harmonics, the PWM waveform of supply voltage is used, set in the general case. Operating characteristics of electric motor drive are set to sub-nominal frequency (f < 50 Hz) and for different angles of the control switching elements in the construction of the inverter. Tensions in the stator of the equivalent machine are determined by three components: the fundamental component; the second component corresponding to the higher order harmonics, which rotate in direct connection; and the appropriate third component of higher order harmonics, which are rotated in reverse. The mathematical model developed will help to determine the harmonics which have a negative influence on the performance of the electric engine and also determines the control angle of the switching elements to remove these harmonics, (Bele 2007).
