Germination response of Hylocereus setaceus (Salm-Dyck ex DC: ) Ralf Bauer (Cactaceae) seeds to temperature and reduced water potentials

The germination response of Hylocereus setaceus seeds to isothermic incubation at different water potentials was analysed by using the thermal time and hydrotime models, aiming to describe some germination parameters of the population and to test the validity of the models to describe the response of the seeds to temperature and water potential. Hylocereus setaceus seeds germinated relatively well in a wide range of temperatures and the germination was rate limited from 11 to 20 °C interval and beyond 30 °C until 40 °C, in which the germination rate respectively shifts positively and negatively with temperature. The minimum or base temperature (Tb) for the germination of H. setaceus was 7 °C, and the ceiling temperature varied nearly from 43.5 to 59 °C depending on the percent fraction, with median set on 49.8 °C. The number of degrees day necessary for 50% of the seeds to germinate in the infra-optimum temperature range was 39.3 °C day, whereas at the supra-optimum interval the value of θ = 77 was assumed to be constant throughout. Germination was sensitive to decreasing values of ψ in the medium, and both the germinability and the germination rate shift negatively with the reduction of ψ, but the rate of reduction changed with temperature. The values of base water potential (ψb) shift to zero with increasing temperatures and such variation reflects in the relatively greater effect of low ψ on germination in supra optimum range of T. In general, the model described better the germination time courses at lower than at higher water potentials. The analysis also suggest that Tb may not be independent of ψ and that ψb(g) may change as a function of temperature at the infra-otimum temperature range.

Somatic development and embryo yield in crossbred F1 mice generated by different mating strategies

The aim of this study was to evaluate different mating strategies among endogamic strains to create F1 populations of mice, minimising the effect of inbreeding depression on somatic development and embryo yield. Females from the strains Swiss, CBA and C57Bl/6 were divided in nine experimental mate arrangements. The total numbers of pups born alive per dam and somatic development, estimated by weighing and measuring the crown-rump length, were recorded. Superovulation response was evaluated in outbreed females. Litter size differed among endogamic dams, irrespective of the sire. Somatic development results suggest heterosis and imprinting phenomena, once a differential parental effect was demonstrated. There was no difference in corpora lutea, ova or embryos recovered (P > 0.05), but recovery and viability rates differ among F1 groups (P < 0.05). The association of dam prolificity with somatic development and superovulation response of the pups should be considered for experimental F1 populations establishment. The use of outbreed animals, however, did not reduce response variability to hormone treatment.

Natural occurrence of the Fusarium solani on Tityus stigmurus (Thorell, 1876) (Scorpiones: Buthidae)

Members of the Fusarium solani species complex are agents of human mycoses, also affecting plants and other animals. Nevertheless, this fungus has not been reported on scorpions. Ten specimens of Tityus stigmurus collected in the field and showing their surface covered by white mycelia were used to assess fungus presence in the animal after its death. Identification of the fungi was based upon the cultural and morphological characteristics. The fungus was isolated from chelicerae and intersegmental regions. Infected individuals had their behaviour modified by reducing feeding and locomotion. None of the infected individuals survived. It is likely that this fungus may have a role in the regulation of field scorpion populations.

Diet of the freshwater stingray Potamotrygon motoro (Chondrichthyes: Potamotrygonidae) on Marajó Island (Pará, Brazil)

The stomach contents of 137 examples of Potamotrygon motoro caught in 3 locations (Muaná, Afuá and Lake Arari) on Marajó Island were analysed. The values of the Index of Relative Importance (IRI) and its respective percentage (%IRI) were calculated. The level of repletion 1 (¼ full) was the most representative for both sexes, as well as for immature and mature specimens. Most of the food items found were well-digested. The food items identification indicated the presence of 15 orders, including insects, mollusks, crustaceans, annelids and fish. Differences in diet were observed among the locations studied when comparing %IRI, crustaceans being the most preferred in Afuá, fish in Lake Arari and mollusks in Muaná.

Mitochondria in the midgut epithelial cells of sugarcane borer parasitized by Cotesia flavipes (Cameron, 1891)

The sugarcane borer Diatraea saccharalis (Lepidoptera: Crambidae) has been controlled by Cotesia flavipes (Hymenoptera: Braconidae); however, very little is known about the effect of the parasitism in the host organs, including the midgut. This work aims to verify mitochondrial alteration in the different midgut epithelial cells of D. saccharalis parasitized by C. flavipes. Midgut fragments (anterior and posterior region) of both non-parasitized and parasitized larvae were processed for transmission electron microscopy. The mitochondria of midgut epithelial cell in the parasitized larvae exhibit morphological alteration, represented by matrix rarefaction and vacuolisation. These mitochondrial alterations are more pronounced in the anterior midgut region during the parasitism process, mainly in the columnar cell.

Water-balance response of Rhinella arenarum (Hensel, 1867) tadpoles to graduated increase in environmental osmolarity

The water balance and the upper limit of osmotic tolerance of premetamorphic Rhinella arenarum larvae (Gosner's stage 26) was evaluated after semistatic incubation in electrolyte (NaCl) and non-electrolyte (mannitol) media following a protocol of progressively increased osmotic pressure. Wet and dry weights were measured to calculate the water content as a derived variable indicative of the hydric balance. Statistical analysis was performed using univariate and integrated multivariate analysis. Tadpoles survived in electrolyte and non-electrolyte solutions up to 200 mOsm. The discriminant function was the best tool to describe the responses of the animals to external environmental stress under experimental conditions. The results were compared with those obtained in previous studies using a protocol of acute exposure to the same media used in this study. It was concluded that a) multivariate analysis is an appropriate approach to describe the responses of tadpoles to changes in the environmental physicochemical parameters, and b) progressive and acute acclimation to the experimental solutions induced similar responses.

Reproduction of the fish community of Passa Cinco Stream, Corumbataí River sub-basin, São Paulo State, Southeastern Brazil

The aim of this work was to evaluate the reproduction (verifying if there was reproductive activity and, if so, with what intensity) of the most abundant species of the fish community in Passa Cinco stream, in relation to the dry and wet season periods and to the longitudinal gradient, through the application of the reproductive intensity index (RII). The sample collection was carried out during one year with six samplings (May, July, September and November 2005, and January and March 2006) in five different points of Passa Cinco stream, contemplating sites of orders two to six. The following fishery equipment was used: a sieve, electric fishery equipment, gill nets and fish-traps. The values of the reproductive intensity index for the dry period were 2.86 and for the wet season 3.17, which indicates the wet season (November to March) as the period when most of the species reproduced. The values of the index for collection point were: 0.78 for site 1, 3.56 for site 2 and 2.89 for site 3. Site 2, which presented the highest value, was the main reproduction place for most of the species. The adults' prevalence in the analysed species suggests, in an isolated way, that those species use the system as a reproduction area. However, when that information was crossed with the values of RII, when intermediate values were considered, it is possible to state that the system is used by the species as much as a reproduction area as a feeding area.

Angiogenic activity of Synadenium umbellatum Pax latex

Synadenium umbellatum Pax, popularly known as "cola-nota", is a medicinal plant that grows in tropical regions. Latex of this plant is used to treat various diseases such as diabetes mellitus, Hansen´s disease, tripanosomiases, leukemia and several malignant tumors. In the present study, the angiogenic activity of S. umbellatum latex was evaluated using the chick embryo chorioallantoic membrane (CAM) assay. Results showed significant increase of the vascular net (p < 0.05) compared to the negative control (H2O). The histological analysis was in accordance with the results obtained. In conclusion, our data indicate that S. umbellatum latex, under the conditions of this research, presented angiogenic effect.

When decisions on homologous structures cause ambiguous taxa relationships: the Neomorphinae (Aves, Cuculidae) example

The anatomy of Neomorphinae is poorly understood and the systematics of this sub-family is also the most controversial of the cuckoo taxa, mainly with regard to the systematic position of Tapera and Dromococcyx. In this study, morphological similarities of the Neomorphinae are discussed after a comprehensive description of the cranial osteology was conducted in seven species, embracing all the Neomorphinae genera. This description is followed by comparisons with other cuckoos in order to contribute to the anatomy and systematics of this sub-family. In this way, we provide illustrations that enable the osteological descriptions and the proposed primary homologies to be visualised and compared. Even though Neomorphinae species share many cranial osteological characteristics, there are some anatomical divergences that allowed us to divide them into two distinct groups: (Dromococcyx/Tapera) and (Morococcyx(Neomorphus/Geococcyx )). After comparisons among all cuckoos this study suggests that Neomorphinae are more similar to Crotophaginae and Couinae than to other sub-families of cuckoos. Our results contrast with a recent phylogenetic study based on morphological features, mainly because alternative interpretations to the primary osteological homologies in this study grouped Tapera and Dromococcyx with Cuculinae. Although morphological studies can be used in phylogenetic analysis, we demonstrated here that decisions in the interpretation of the homologies can provide ambiguous results.

Isolation and experimental infection with Vibrio alginolyticus in the sea horse, Hippocampus reidi Ginsburg, 1933 (Osteichthyes: Syngnathidae) in Brazil

The aim of this study was to evaluate the pathogenicity of Vibrio alginolyticus isolated from an outbreak of sea horse Hippocampus reidi reared in the State of Santa Catarina, Brazil, by experimental infection. Sea horses with necrosis on the mouth epithelium were collected from aquaria at the Aquaculture Department, UFSC and the bacterium isolated from the mouth, liver, heart and blood in tiosulphate citrate bilesalt sucrose agar broth. The strains were identified by API 20E kit with 99.1% probability as Vibrio alginolyticus. Twelve adult sea horses (9.63 ± 2.42 g and 15.12 ± 0.87 cm) were distributed in six aquaria of 10 L capacity with aerated sea water. Fish from three aquaria were submitted to an immersion bath in a solution containing 1.0 × 10(7) CFU of V. alginolyticus/mL for 15 minutes. Fish from the other three aquaria received the same procedure without bacteria. Twenty four hours after this challenge, 100% mortality was observed in the animals infected with V. alginolyticus. No mortality was observed in non-infected fish. Hyperplasia, displacement and fusion of secondary lamellae of the gills; leukocyte infiltration and necrotic foci in the kidney; hyperplasia, sinusoidal deformation and necrotic foci in the liver were observed in histopathological analysis. The V. alginolyticus isolated in this study was pathogenic to H. reidi and constitutes an important sanitary problem to its culture.

Acoustic communication and vocalization microhabitat in Ameerega braccata (Steindachner, 1864) (Anura, Dendrobatidae) from Midwestern Brazil

Ameerega braccata is an aposematic, small dendrobatid anuran known from its type-locality, Chapada dos Guimarães, in the State of Mato Grosso, and from a few additional localities in Mato Grosso do Sul and Goiás States, Brazil. The advertisement call of A. braccata is composed of a single unpulsed note, with a frequency range from 3.5 to 4.2 KHz (N = 110), and average duration of 65.8 ms (N = 110, SD = 11.6). The territorial call is composed of five or six repeated notes, structurally similar to advertisement call notes. The courtship call is emitted in close-range interactions between male-female during the courtship event and may reach frequencies of 2.2 to 5.3 KHz (N = 10), with shorter notes (average duration 43 ms; N = 10; SD = 4.9). Call duration, note duration and call rate of the advertisement call showed high variation (>15% of coefficient of variation), and dominant frequency showed low variation (<4%). The properties with higher variation possibly respond to climatic features and male social position. When producing advertisement calls, males were usually found at an average height of 31.4 cm (N = 19, SD = 12.2 cm), mainly on leaves of shrubs and herbaceous plants (59%). Individuals of A. braccata were found in open physiognomies in the Cerrado ("campo sujo" and "cerrado stricto sensu"), as opposed to that found by previous researchers, who reported an association of the species with gallery forests in the Cerrado.

Histological study of the dynamics in epidermis regeneration of the carp tail fin (Cyprinus carpio, Linnaeus, 1758)

Teleostean fins when partially amputated suffer a regenerative process called epimorphic regeneration, characterized by the following stages: healing, based on the formation of a multistratified epidermal layer, the formation of a mass of pluripotent cells known as blastema, the differentiation of these cells, the synthesis and disposition of the extracellular matrix, morphological growth and restoration. The epidermis has a fundamental role in the regenerative process of fish fins, as the healing time of this structure leads it to a faster regenerative process and it also works as a defense against the external environment. In this sense, due to the fast regeneration shown by the epidermis, the aim of this paper is to study the histology of the regenerative dynamics of the carp fin tail (Cyprinus carpio), under the light and transmission electron microscope. Epidermic regeneration begins right in the first hours after the fin amputation and it continues throughout the regenerative process. After 24 hours, an apical epidermal cap is established. Cytoplasmatic prolongations and intercellular junctions are observed and the cells of the basal layer of the epidermis change from the cubic form to the cylindrical, due to the development of the cytoplasmatic organelles responsible for the synthesis of the basal membrane, lost after amputation. These results show the importance of histological studies in regenerative processes. We believe that the association of molecular biology with histological studies can throw further light onto these regenerative dynamics.

New data on the ecology and geographic distribution of Saguinus inustus Schwarz, 1951 (Primates, Callitrichidae)

Saguinus inustus (Schwarz, 1951) is one of the neotropical primates least studied. The distribution of the species ranges from the north of the Solimões River, between the Negro and Japurá Rivers in Brazil, and Guayabero-Guaviare Rivers in Colombia. Nevertheless, due to the low number of specimens collected from the lower Japurá and lower Negro Rivers areas, the geographic distribution is so far poorly delineated. In this study, field data was composed of sightings and the collection of specimens during a survey of mammal diversity in the Amana Sustainable Development Reserve (ASDR). For this survey, two 40-day expeditions were carried out in 2004. The first one occurred during the flooded season in June and July, and the second was during the peak of the dry season in October. Direct sightings were made through hiking along transects, navigation along water channels with a 30-hp speedboat, and gliding along flooded trails in the forest. New records of S. inustus were made in 11 different localities in ASDR. The study has confirmed the presence of the species in the Amanã area, carrying out the first records of the species in flooded forest habitats.

Effects of fishing technique on assessing species composition in aquatic systems in semi-arid Brazil

In most ecological field research, appropriate sampling is critical for the understanding of processes underlying fish populations and communities, and is even more important in heterogeneous environments such as the aquatic systems of the semi-arid region of Brazil. This study intends to make a contribution to the development of sampling programs and gear selection in aquatic systems of semi-arid Brazil by evaluating the effects of different fishing techniques on the assessment of richness and composition of the fish fauna in selected aquatic environments. Six sites were selected to represent typical artificial (reservoirs) and natural (intermittent streams) environments and four different types of sampling gear were applied to each site during four occasions. The present study shows that when selecting sampling techniques to be used in aquatic systems in semi-arid Brazil, one must consider the objectives of the study, e.g. ecological or taxonomic, in order to decide on inclusion of rare species in the sampling population. Also, the effect of the sampling gear on natural abundances of fish must be considered given that some sampling techniques are highly detrimental to fish population numbers.

How many studies are necessary to compare niche-based models for geographic distributions? Inductive reasoning may fail at the end

The use of ecological niche models (ENM) to generate potential geographic distributions of species has rapidly increased in ecology, conservation and evolutionary biology. Many methods are available and the most used are Maximum Entropy Method (MAXENT) and the Genetic Algorithm for Rule Set Production (GARP). Recent studies have shown that MAXENT perform better than GARP. Here we used the statistics methods of ROC - AUC (area under the Receiver Operating Characteristics curve) and bootstrap to evaluate the performance of GARP and MAXENT in generate potential distribution models for 39 species of New World coral snakes. We found that values of AUC for GARP ranged from 0.923 to 0.999, whereas those for MAXENT ranged from 0.877 to 0.999. On the whole, the differences in AUC were very small, but for 10 species GARP outperformed MAXENT. Means and standard deviations for 100 bootstrapped samples with sample sizes ranging from 3 to 30 species did not show any trends towards deviations from a zero difference in AUC values of GARP minus AUC values of MAXENT. Ours results suggest that further studies are still necessary to establish under which circumstances the statistical performance of the methods vary. However, it is also important to consider the possibility that this empirical inductive reasoning may fail in the end, because we almost certainly could not establish all potential scenarios generating variation in the relative performance of models.

Spatial patterns of a tropical tree species growing under an eucalyptus plantation in South-East Brazil

The objectives of this study were to evaluate the influence of propagules source and the implication of tree size class on the spatial pattern of Xylopia brasiliensis Spreng. individuals growing under the canopy of an experimental plantation of eucalyptus. To this end, all individuals of Xylopia brasiliensis with diameter at soil height (dsh) > 1 cm were mapped in the understory of a 3.16 ha Eucalyptus spp. and Corymbia spp. plantation, located in the municipality of Lavras, SE Brazil. The largest nearby mature tree of X. brasiliensis was considered as the propagules source. Linear regressions were used to assess the influence of the distance of propagules source on the population parameters (density, basal area and height). The spatial pattern of trees was assessed through the Ripley K function. The overall pattern showed that the propagules source distance had strong influence over spatial distribution of trees, mainly the small ones, indicating that the closer the distance from the propagules source, the higher the tree density and the lower the mean tree height. The population showed different spatial distribution patterns according to the spatial scale and diameter class considered. While small trees tended to be aggregated up to around 80 m, the largest individuals were randomly distributed in the area. A plausible explanation for observed patterns might be limited seed rain and intra-population competition.

Coliform accumulation in Amphibalanus amphitrite (Darwin, 1854) (Cirripedia) and its use as an organic pollution bioindicator in the estuarine area of Recife, Pernambuco, Brazil

Samples of water and barnacles Amphibalanus amphitrite were collected from Recife, Brazil, to assess if it accumulates total (TC) and thermotolerant coliforms (TTC) related with sewage pollution. The Most Probable Number (MPN) values and the standard procedures for examination of shellfish were used. Comparatively with the water samples, the highest coliform values came from the barnacles, with TC values ranging from < 3.0 × 10³ to > 2.4 × 10(6) MPN.g-1, and TTC ranging from > 2.4 × 10³ to 2.9 × 10(5) MPN.g-1. Barnacles accumulate the TC Ewingella americana, and the TTC Escherichia coli, Enterobacter gergoviae, Enterobacter aerogenes, and Enterobacter sakazakii. The results provided an indication of the level of organic contamination at the sampling locations and that this species could be a good organic pollution bioindicator.

Interference of Pteridium arachnoideum (Kaulf.) Maxon. (Dennstaedtiaceae) on the establishment of rainforest trees

In order to identify the effect of P. arachnoideum, we studied 11 native tree species commonly used in reforestation projects. Bioassays were conducted in laboratory to evaluate the effect of bracken leachate on the germination and morphology of seedlings. Juveniles of some of these species were planted in two adjacent but contrasting areas in relation to the dominance of P. arachnoideum. The evaluation of growth and survivorship was performed after six and twelve months. This study reveals that for some pioneer and secondary trees P. arachnoideum leachate exerted an inhibitory effect on seed germination and seedling morphology. Field experiments revealed that pioneers are apparently more resistant to P. arachnoideum leachate than secondary species.

Q10 of heterotrophic activity during aerobic decomposition of Utricularia breviscapa and its effect on carbon cycling in a tropical lagoon

In this study the Q10 coefficients of heterotrophic activities were measured during aerobic decomposition of Utricularia breviscapa Wright ex Griseb from Óleo lagoon (21° 36' S and 49° 47' W), Luiz Antonio, SP. The bioassays were set up with fragments of U. breviscapa and incubated with lagoon water at distinct temperatures (15.3, 20.8, 25.7 and 30.3 °C). Periodically for 95 days, the concentrations of dissolved oxygen were determined in the bioassays. The results of the temporal variation of dissolved oxygen were fitted to a first-order kinetic model. The stoichiometric relations were calculated on the basis of these fittings. In general, the results allowed us to conclude: i) the oxygen/carbon stoichiometric relations (O/C) varied in function of temperature and time. The temporal variations of the O/C observed in the decomposition of U. breviscapa, suggest that, in the initial phases of the process, low organic carbon concentrations were enough to generate great demands of oxygen, ii) the oxygen consumption coefficients (k d) presented low variation in function of increasing temperature, iii) the increment of the temperature induced a higher consumption of oxygen (COmax) and iv) the simulations indicate that during summer, temperature activates the metabolism of decomposing microbiota.

Straminipilous organisms growing on herbivorous pirapitinga (Piaractus brachypomus) and carnivorous piranha (Pygocentrus nattereri) from Poland

We investigated the growth of straminipilous organisms on the skin, muscles and liver of herbivorous pirapitinga (Piaractus brachypomus) and carnivorous piranha (Pygocentrus nattereri) in water of three different eutrophication levels. Sixteen straminipilous organism species were found growing on the investigated body parts of both species of fish used as baits. The higher number of species was found on the baits of carnivorous species (15) when compared with the ones from the herbivorous pirapitinga (10 species). The highest number of straminipilous organisms species developed on the skin of both species of fish. The highest number of species of straminipilous organisms was observed growing in the water of the BiaBa river (middle eutrophication), while the lowest number occurred in the baits of vessels with water from the Dojlidy pond (low eutrophication).
