SEM and 3D microtomography application to investigate the distribution of fibers in advanced cementitious composites
Abstract The incorporation of fibers on cement composites reduces its fragility, turning it a ductile material. The amount of fibers and its distribution presents a large influence on the composite performance, especially by the fact that it is self-consolidating, which facilitates the fiber distribution. 3D microtomography is an efficient tool for determining the fibers distribution, generating images and creating a representation in three dimensions of the sample. Moreover, the scanning electron microscopy (SEM) can be used to analyze the interaction of fibers with the cement composite. The purpose of this paper is to investigate the application of these techniques to visualize the distribution and interaction of metallic and polypropylene fibers inserted into an advanced cementitious composite, at 3% in volume content. The results presented these techniques' efficiency in the verification of fibers distribution within the mixture and the absence of flaws in the composition.

Influence of the flexibility of beams and slabs in static response and dynamic properties
Abstract This article examines numerically the flexibility influence of support beams in static response and dynamic properties of a symmetric plate formed by massive slabs of reinforced concrete in elastic linear regime, using the Finite Element Method. In the static response the variation of bending mo-ments and displacements are evaluated, which depend on the relationship between the flexibility of the slab and the beam. The evaluation of dynamic properties is held in undamped free vibration, through which the vibration modes and the values of the natural frequencies is obtained, which are compared with the limits of the Brazilian standard code for design of concrete structures. Results show that the response may show great variation due to the change in the relationship between bending stiffness of the slabs and the beams.

The influence of connecting pile cap-column in the mechanisms of break in the two pile caps
Abstract The paper analyzes the two pile caps with partially embedded socket and subject a center load. Three models were experimentally tested, varying the type of conformation of the column and walls of the socket, with a smooth, the other rough, and a monolithic two pile cap, used for reference. The roughening of the column-socket interface was examined with the aim of verifying the difference of the distribution of compressive and tensile stresses in the strut an tie model used for design. The experimental test to show that the two pile caps with conformation rough of the column and walls of the socket, support more load in comparison with two pile caps with smooth of the column and walls of the socket. Both however underperformed the monolithic two pile cap, with values of 66% and 36% respectively.

Effect of wind in the design of reinforced concrete buildings
ABSTRACT This paper presents the results from a parametric study carried in order to quantify how far errors in the design stage related to the consideration of the wind action may put at risk the response and safety of reinforced concrete buildings. Using an architectural model as reference and varying the number of floors of the building, the structural safety was evaluated as a function of the wind action intensity. Results showed that even for low-rise buildings, with 10 floors, ignoring the wind action can significantly jeopardize their behaviour and safety. Yet, for slenderer buildings, up to 30 floors, it can lead to catastrophic results, as the ruin of the structure by progressive collapse.

Performance evaluation of plastic spacers: proposal and development of evaluation methods
ABSTRACT The durability of reinforced concrete structures is highly dependent on the characteristics of the concrete cover to reinforcement and its thickness. The failure to obtain cover thickness is the factor with the largest influence on the premature corrosion of the reinforcement, which in turn is the main deterioration form of reinforced concrete structures. Studies indicate that the designed cover is not reached in the current constructions that adopt this structural solution, configuring a chronic, and not a sporadic problem. One of the observed causes for the failure in obtaining the minimum standardized cover is the incorrect use of spacers and the use of inadequate spacers. This is made more serious by the absence of a Brazilian standard to regulate the product and its use and, consequently, the absence of a quality certification from the responsible regulating agency. Focusing on spacers, requirements and performance criteria were proposed, in addition to methods for their assessment, with most being taken and adapted from international standards. Subsequently, some spacers available on the market were effectively tested according to the proposed methodology. No spacer model proved to be satisfactory according to the established performance approach. However, for each criteria and assessment methods proposed, there was, at least, one spacer model at the market which satisfied them, so it can be said that the criteria and methods are suitable for spacers performance evaluation. Faced with the performance diversity of the spacer models on offer, the need for a regulatory Brazilian standard for this product was confirmed in order to delimit the quality of spacers available on the market and to eliminate this variable as one of the causes for not obtaining the correct covering.

Temperature monitoring in large volume spread footing foundations: case study "Parque da Cidade" - São Paulo
ABSTRACT In recent years, the construction of foundation elements from large-volume reinforced concrete is becoming increasingly common. This implies a potential increase in the risk of cracks of thermal origin, due to the heat of hydration of cement. Under these circumstances, these concrete elements need to be treated using the mass concrete theory, widespread in dam construction, but little used when designing buildings. This paper aims to present a case study about the procedures and problems involved in the construction of a spread footing with a volume of approximately 800m³ designed for the foundation of a shopping center in São Paulo, Brazil.

Evaluating the applicability of rheometry in steel fiber reinforced self-compacting concretes
ABSTRACT The use of self-compacting concrete in civil construction industry presents various advantages, since the material shows adequate workability during fresh state. When fiber reinforcement is used, there are changes in its behavior that require attention. This study aimed to evaluate the applicability of rheological tests and the correlation between its results and those obtained with regular tests used to control SCCs. In that sense, different mixtures of SCC with different steel fiber contents were produced in order to be analyzed in the experimental program described. Rotational rheology tests and slump flow and L-box tests were performed. The results showed that slump flow test did not present good correlations with rheological parameters. On the other hand, this test was able to point out the risk of segregation of the mixtures with higher fiber contents. A good correlation was obtained between the L-box test results and rheological parameters. The L-box was also able to show loss in the passing ability of SCC related to the rise of plastic viscosity and yield stress. These tests also presented a good correlation with the rheological parameters when lower fiber contents were used. Although the rheometry test was unable to evaluate these problems, this kind of test gave more objective and reliable data on variations in rheological parameters related to the increase of fiber content, and proved to be a more reliable test to this kind of application, especially when applied together with the conventional tests.

Sewage sludge ash (SSA) in high performance concrete: characterization and application
ABSTRACT Sewage sludge originated from the process of treatment of wastewater has become an environmental issue for three main reasons: contains pathogens, heavy metals and organic compounds that are harmful to the environmental and human health; high volumes are daily generated; and shortage of landfill sites for proper disposal. This research deals with the viability study of sewage sludge utilization, after calcination process, as mineral admixture in the production of concrete. High-performance concretes were produced with replacement content of 5% and 10% by weight of Portland cement with sewage sludge ash (SSA). The influence of this ash was analyzed through physical and mechanical tests. Analysis showed that the mixtures containing SSA have lower values of compressive strength than the reference. The results of absorptivity, porosity and accelerated penetration of chloride ions, presents that mixtures containing ash showed reductions compared to the reference. This indicates that SSA provided refinement of the pore structure, which was confirmed by mercury intrusion porosimetry test.

Treatment with mebendazole is not associated with distal migration of adult Angiostrongylus costaricensis in the murine experimental infection
Abdominal angiostrongyliasis is a zoonotic infection produced by a metastrongylid intra-arterial nematode, Angiostrongylus costaricensis. Human accidental infection may result in abdominal lesions and treatment with anti-helminthics is contra-indicated because of potential higher morbidity with excitement or death of worms inside vessels. To evaluate the effect of mebendazole on localization of the worms, male Swiss mice, 5 week-old, were infected with 10 third stage larvae per animal. Twelve infected mice were treated with oral mebendazol, at 5 mg/kg/day, for 5 consecutive days, begining 22 days after inoculation. As control groups, 12 infected but non-treated mice and other 12 non-infected and non-treated mice were studied. The findings at necropsy were, respectively for the treated (T) and control (C) groups: 92% and 80% of the worms were inside the cecal mesenteric arterial branch; 8% and 10% were located inside the aorta. Only in the group C some worms (10%) were found inside the portal vein or splenic artery. These data indicate that treatment with mebendazole does not lead to distal or ectopic migration of A. costaricensis worms.

Study of enteroparasites infection frequency and chemotherapeutic agents used in pediatric patients in a community living in Porto Alegre, RS, Brazil
Parasitic infections caused by intestinal protozoan and helminths affect more than two billion people worldwide and chemotherapy is the most commonly used therapeutic procedure. Considering the problems created by parasitic infections and the incorrect use of drugs, the aim of this work was to detect the frequency of enteroparasites infection and to estimate the use of chemotherapeutic agents in children living in the periphery of the city of Porto Alegre, RS, Brazil. Ninety-six preschool age children, who had parasitological exams and who used antiparasitic drugs, were analyzed. The efficacy of treatment was evaluated by stool examination repeated six months after treatment. The same diagnostic test was used to evaluate parasitological cure, which was defined as absence of eggs and cysts in the stool. From these children, 79 (82.3%) were contaminated by some species of parasite, the most prevalent were Ascaris lumbricoides, Trichuris trichiura and Giardia lamblia. The most commonly used drugs were mebendazole (86% of prescriptions) and metronidazole (30.3%). The cure rate in the 79 children, examined 6 months after treatment, was 65.3% for A. lumbricoides and 66.1% for T. trichiura. This study suggests that a continuous education program regarding the prevention and treatment of parasitic infections is an essential tool for their eradication.

Toxocaríase humana: freqüência de anticorpos anti-Toxocara em crianças e adolescentes atendidos em um ambulatório especializado de filariose linfática em Recife, Nordeste do Brasil
In a transversal study on a sample of 386 children and adolescents from an outpatient clinic for filariasis in Recife, Northeast Brazil, the frequency of anti-Toxocara antibodies and its relation to age, gender, number of peripheral eosinophils, Wuchereria bancrofti microfilariae and intestinal helminths was determined. The total anti-Toxocara IgG antibody frequency was 39.4%, by ELISA technique. The difference in frequency between males (40.1%) and females (37.6%) was not statistically significant. The 6 to 10-year-old subset presented the highest frequency of anti-Toxocara antibodies (60%), and within this age group there was a statistically significant male bias. There was also a significant association between the number of eosinophils and the presence of anti-Toxocara antibodies. Intestinal parasite frequency was 52.1%, but no association was found between this data and the presence of anti-Toxocara antibodies. In the present sample, 42.2% of the patients were Wuchereria bancrofti carriers, however, again this was not associated with the presence of anti-Toxocara antibodies. In conclusion, anti-Toxocara antibodies were highly prevalent in this sample. The present data show that there is no cross correlation between anti-Toxocara IgG antibody and the presence of intestinal helminths and filariasis.

Determination of human cytomegalovirus genetic diversity in different patient populations in Costa Rica
Seroprevalence of HCMV in Costa Rica is greater than 95% in adults; primary infections occur early in life and is the most frequent congenital infection in newborns. The objectives of this study were to determine the genetic variability and genotypes of HCMV gB gene in Costa Rica. Samples were collected from alcoholics, pregnant women, blood donors, AIDS patients, hematology-oncology (HO) children and HCMV isolates from neonates with cytomegalic inclusion disease. A semi-nested PCR system was used to obtain a product of 293-296 bp of the gB gene to be analyzed by Single Stranded Conformational Polymorphism (SSCP) and sequencing to determine the genetic polymorphic pattern and genotypes, respectively. AIDS patients showed the highest polymorphic diversity with 14 different patterns while fifty-six percent of HO children samples showed the same polymorphic pattern, suggesting in this group a possible nosocomial infection. In neonates three genotypes (gB1, gB2 and gB3), were determined while AIDS patients and blood donors only showed one (gB2). Of all samples analyzed only genotypes gB1, 2 and 3 were determined, genotype gB2 was the most frequent (73%) and mixed infections were not detected. The results of the study indicate that SSCP could be an important tool to detect HCMV intra-hospital infections and suggests a need to include additional study populations to better determine the genotype diversity and prevalence.

Childhood astrovirus-associated diarrhea in the ambulatory setting in a Public Hospital in Cordoba city, Argentina
Human astroviruses have been increasingly identified as important agents of diarrheal disease in children. However, the disease burden of astrovirus infection is still incompletely assessed. This paper reports results on the epidemiological and clinical characteristics of astrovirus-associated diarrhea, as well as the impact of astrovirus infection on the ambulatory setting at a Public Hospital in Córdoba city, Argentina. From February 2001 through January 2002, 97 randomly selected outpatient visits for diarrhea among children < 36 months old were enrolled. A single specimen of stool from each child was collected and tested for astrovirus antigen by enzyme immunoassay. Astroviruses were detected in 12.37% of the diarrheal episodes. All the positive cases occurred in children 4 to 18 months, but the highest rate was in children aged 4 to 6 months (23.80%). The clinical symptoms of astrovirus associated-diarrhea were fever 41.66%, vomiting 25.00% and dehydration 8.33%; overall 16.66% required hospitalization. Astrovirus was identified through the year and no seasonally pattern was detected (cool semester 15.21% versus warm semester 9.80% p >; 0.05). According to our estimation about one out of seventy-four children in this cohort would be assisted annually for an astroviral-diarrheal episode in the Public Hospital and one out of eight diarrheal cases could be attributed to astrovirus infection. Astrovirus is a common symptomatic infection in pediatric outpatient visits in the public hospital in the study area, contributing 12.37% of the overall morbidity from diarrhea.

Effects of eucalyptol on house fly (Diptera: Muscidae) and blow fly (Diptera: Calliphoridae)
The effects of eucalyptol were evaluated against the house fly, Musca domestica L., and blow fly, Chrysomya megacephala (F.). The bioassay of adults, using topical application, indicated that M. domestica males were more susceptible than females, with the LD50 being 118 and 177 µg/fly, respectively. A higher LD50 of C. megacephala was obtained; 197 µg/fly for males and 221 µg/fly for females. Living flies of both species yielded a shorter life span after being treated with eucalyptol. The bioassay of larvae, using the dipping method on the third instar, showed that M. domestica was more susceptible than C. megacephala, with their LC50 being 101 and 642 µg/µl, respectively. The emergence of adults, which had been treated with eucalyptol in larvae, decreased only in M. domestica. Having the volatile property, fumigation or impregnated paper test of eucalyptol or the efficacy of repellence or attractiveness merits further investigations to enhance bio-insecticidal efficacy.

Immunogenicity of three recombinant hepatitis B vaccines administered to students in three doses containing half the antigen amount routinely used for adult vaccination
We evaluated the immunogenicity of three recombinant hepatitis B vaccines, one Brazilian (Butang, Instituto Butantan) and two Korean vaccines (Euvax-B, LG Chemical Ltd. and Hepavax-Gene, Greencross Vaccine Corp.), administered intramuscularly to students aged 17 to 19 years in three 10-µg doses (corresponding to half the amount of antigen routinely used for adult vaccination) at intervals of one month between the first and second dose, and of four months between the second and third dose. A total of 316 students non-reactive for any serological marker of hepatitis B virus infection were vaccinated: 77 (24.4%) with the Butang vaccine, 71 (22.5%) with Euvax-B, 85 (26.9%) with Hepavax-Gene and, for comparison, 83 (26.2%) with Engerix-B (GlaxoSmithKline), whose efficacy in young adults at the dose used here has been confirmed in previous studies. Similar seroconversion rates (anti-HBs >; 10 mIU/mL about one month after application of the third dose) were obtained for the Butang, Euvax-B, Hepavax-Gene and Engerix-B vaccines (96.2%, 98.6%, 96.5% and 97.6%, respectively). The frequency of good responders (anti-HBs >; 100 mIU/mL) was also similar among students receiving the four vaccines (85.8%, 91.6%, 89.4% and 89.2%, respectively). The geometric mean titers (GMT) of anti-HBs about one month after the third dose obtained with these vaccines were 727.78 ± 6.46 mIU/mL, 2009.09 ± 7.16 mIU/mL, 1729.82 ± 8.85 mIU/mL and 2070.14 ± 11.69 mIU/mL, respectively. The GMT of anti-HBs induced by the Euvax-B and Engerix-B vaccines were higher than those obtained with the Butang vaccine (p < 0.05); this difference was not significant when comparing the other vaccines two-by-two. No spontaneous adverse effects attributable to the application of any dose of the four vaccines were reported.

Mapeamento de um gene de Leishmania major que confere resistência a pentamidina por deleção e inserção de elementos transposicionais
Pentamidine (PEN) is an alternative compound to treat antimony-resistant leishmaniasis patients, which cellular target remains unclear. One approach to the identification of prospective targets is to identify genes able to mediate PEN resistance following overexpression. Starting from a genomic library of transfected parasites bearing a multicopy episomal cosmid vector containing wild-type Leishmania major DNA, we isolated one locus capable to render PEN resistance to wild type cells after DNA transfection. In order to map this Leishmania locus, cosmid insert was deleted by two successive sets of partial digestion with restriction enzymes, followed by transfection into wild type cells, overexpression, induction and functional tests in the presence of PEN. To determine the Leishmania gene related to PEN resistance, nucleotide sequencing experiments were done through insertion of the transposon Mariner element of Drosophila melanogaster (mosK) into the deleted insert to work as primer island. Using general molecular techniques, we described here this method that permits a quickly identification of a functional gene facilitating nucleotide sequence experiments from large DNA fragments. Followed experiments revealed the presence of a P-Glycoprotein gene in this locus which role in Leishmania metabolism has now been analyzed.

The frequency rate of Toxocara species contamination in soil samples from public yards in a urban area "Payathai", Bangkok, Thailand
Toxocara species are most common roundworms of Canidae and Felidae. Human toxocariasis develops by ingesting of embryonated eggs in contaminated soil. There is no previous report of Toxocara contamination in the soil samples from the public areas in Bangkok. For this reason our study have been carried out to examine the frequency of Toxocara eggs in public yards in Bangkok, Thailand. A total of 175 sand and clay samples were collected and examined for parasite eggs. According to this study, Toxocara eggs were detected from 10 (5.71%) of 175 soil samples. The high rate of contamination in this study implies the importance of the control of this possible zoonotic disease: control of abandon of dogs and cats, is still necessary.

"Multiplex PCR" identification of the atypical and monophasic Salmonella enterica subsp. enterica serotype 1,4,[5],12:i:- in São Paulo State, Brazil: frequency and antibiotic resistance patterns
Salmonella spp. are the etiologic agents of salmonellosis, a worldwide spread zoonoses causing foodborne outbreaks and clinical diseases. By serological identification, Salmonella enterica subsp. enterica serotype 1,4,[5],12:i:- accounted for 8.8% of human and 1.6% of nonhuman Salmonella strains isolated in São Paulo State, during 1991-2000. A total of 28.6% of them amplified a fragment corresponding to H:1,2 (flagellar phase two) through PCR analysis and were further assigned as S. Typhimurium. Antimicrobial resistance was detected in 36.3% of the 369 PCR-negative strains tested, including the multiresistance to ampicillin, chloramphenicol, sulfonamides, tetracycline, and streptomycin.
