Effects of acute and chronic physical exercise and stress on different types of memory in rats

Here we study the effect of acute and chronic physical exercise in a treadmill and of daily stress (because forced exercise involves a degree of stress) during 2 or 8 weeks on different types of memory in male Wistar rats. The memory tests employed were: habituation in an open field, object recognition and spatial learning in the Morris water maze. Daily foot-shock stress enhanced habituation learning after 2 but not after 8 weeks; it hindered both short- (STM) and long-term memory (LTM) of the recognition task at 2 weeks but only STM after 8 weeks and had no effect on spatial learning after either 2 or 8 weeks. Acute but not chronic exercise also enhanced habituation in the open field and hindered STM and LTM in the recognition task. Chronic exercise enhanced one important measure of spatial learning (latency to escape) but not others. Our findings indicate that some care must be taken when interpreting effects of forced exercise on brain parameters since at least part of them may be due to the stress inherent to the training procedure.

Limited replication of yellow fever 17DD and 17D-Dengue recombinant viruses in rhesus monkeys

For the development of safe live attenuated flavivirus vaccines one of the main properties to be established is viral replication. We have used real-time reverse transcriptase-polymerase chain reaction and virus titration by plaque assay to determine the replication of yellow fever 17DD virus (YFV 17DD) and recombinant yellow fever 17D viruses expressing envelope proteins of dengue virus serotypes 2 and 4 (17D-DENV-2 and 17D-DENV-4). Serum samples from rhesus monkeys inoculated with YFV 17DD and 17D-DENV chimeras by intracerebral or subcutaneous route were used to determine and compare the viremia induced by these viruses. Viral load quantification in samples from monkeys inoculated by either route with YFV 17DD virus suggested a restricted capability of the virus to replicate reaching not more than 2.0 log10 PFU mL-1 or 3.29 log10 copies mL-1. Recombinant 17D-dengue viruses were shown by plaquing and real-time PCR to be as attenuated as YF 17DD virus with the highest mean peak titer of 1.97 log10 PFU mL-1 or 3.53 log10 copies mL-1. These data serve as a comparative basis for the characterization of other 17D-based live attenuated candidate vaccines against other diseases.

The status of the name Alysium holtingii C. Agardh, a red alga described from Brazil, and a depiction of the type specimen

The type specimen of the red alga Alysium holtingii C. Agardh, described from Brazil, is located in the Lund Herbarium, and it is depicted for the first time in a publication. It is taxonomically identical to Dichotomaria obtusata (J. Ellis and Solander) Lamarck and thus can be treated as a later taxonomic synonym. Alysium is regarded as congeneric with Dichotomaria.

Molluscicidal activity of 2-hydroxy-[1,4]naphthoquinone and derivatives

The toxic profile of lawsone (2-hydroxy-[1,4]naphthoquinone) and a series of [1,4]naphthoquinone derivatives was evaluated against the brine shrimp Artemia salina and against the mollusk Biomphalaria glabrata, the main transmitting vector of schistosomiasis in Brazil. Of the seventeen compounds tested nine fell below the threshold of 100 µg/mL set for potential molluscicidal activity by the World Health Organization. As a general rule derivatives with non-polar substituents presented the highest molluscicidal activities. These substances showed significant toxicity in A. salina lethality bioassay.

The influence of bromazepam on cortical power distribution

The EEG has been widely employed in the assessment of electrophysiological changes induced by distinct medications. Its sensibility in detecting alterations produced by a specific substance may be enhanced by methods of quantitative analyses (qEEG). The present study aimed at investigating the modulatory effects of bromazepam on brain dynamics. The effects of bromazepam (3mg) on EEG power distribution were tested in 10 healthy individuals, in a double-blind experiment. The electrophysiological measure was analyzed across experimental conditions, moments, and electrodes, in the delta, theta, alpha and beta frequency bands separately. A significant decrease of relative power was observed in delta and theta (main effect of condition). No interactions were observed. Although the expected anxiolytic EEG profile was not observed (increased beta and decreased alpha activity), this specific result may be related to other factors such as dosage used and the subjects' general physiological state, and not necessarily to the drug itself.

A Holocene pollen record of savanna establishment in coastal Amapá

The main goal of this study was to investigate how climate and human activities may have influenced ecotonal areas of disjoint savannas within Brazilian Amazonia. The fossil pollen and charcoal records of Lake Márcio (Amapá) were used to provide a Holocene palaeoecological history of this region. Detrended correspondence analysis (DCA) was used to enhance the patterns of sample distribution along the sediment core. A marked vegetation change from closed forests with swamp elements to open flooded savanna at c. 5000 yrs BP was evident from the pollen record. Charcoal analysis revealed a pattern of increased accumulation of particles coincident with the establishment of savannas, suggesting higher fire frequency and human impacts near the lake. A 550-year sedimentary hiatus suggests that the lake depended heavily on floodwaters from the Amazon River, and that it became suddenly isolated from it. When sedimentation restarted in the lake, the environment had changed. A combination of factors, such as reduced river flooding, palaeofires and human occupation may have had a tremendous impact on the environment. As there are no other major changes in vegetation, after 4700 yrs BP, it is plausible to assume that the modern mosaic vegetation formed at that time.

Geochemistry and petrogenesis of post-collisional ultrapotassic syenites and granites from southernmost Brazil: the Piquiri Syenite Massif

The Piquiri Syenite Massif, southernmost Brazil, is part of the post-collisional magmatism related to the Neoproterozoic Brasiliano-Pan-African Orogenic Cycle. The massif is about 12 km in diameter and is composed of syenites, granites, monzonitic rocks and lamprophyres. Diopside-phlogopite, diopside-biotite-augite-calcic-amphibole, are the main ferro-magnesian paragenesis in the syenitic rocks. Syenitic and granitic rocks are co-magmatic and related to an ultrapotassic, silica-saturated magmatism. Their trace element patterns indicate a probable mantle source modified by previous, subduction-related metasomatism. The ultrapotassic granites of this massif were produced by fractional crystallization of syenitic magmas, and may be considered as a particular group of hypersolvus and subsolvus A-type granites. Based upon textural, structural and geochemical data most of the syenitic rocks, particularly the fine-grained types, are considered as crystallized liquids, in spite of the abundance of cumulatic layers, schlieren, and compositional banding. Most of the studied samples are metaluminous, with K2O/Na2O ratios higher than 2. The ultrapotassic syenitic and lamprophyric rocks in the Piquiri massif are interpreted to have been produced from enriched mantle sources, OIB-type, like most of the post-collisional shoshonitic, sodic alkaline and high-K tholeiitic magmatism in southernmost Brazil. The source of the ultrapotassic and lamprophyric magmas is probably the same veined mantle, with abundant phlogopite + apatite + amphibole that reflects a previous subduction-related metasomatism.

In-flight absolute calibration of the CBERS-2 CCD sensor data

Since the first images of the sensors on board of CBERS-2 (China-Brazil Earth Resources Satellite) satellite were made available by the National Institute for Space Research (INPE), users have asked information about the conversion of image digital numbers to physical data (radiance or reflectance). This paper describes the main steps that were carried out to calculate the in-flight absolute calibration coefficients for CBERS-2 CCD level 2 (radiometric and geometric correction) images considering the reflectance-based method. Remarks about the preliminary evaluation of these coefficients application are also presented.

Study of wet precipitation and its chemical composition in South of Brazil

The purpose of this study is to analyze the chemical composition of wet precipitation in samples collected at three stations in the Candiota region in the Brazilian state of Rio Grande do Sul (RS). Samples were collected in 2004. Variables analyzed in wet precipitation were pH, conductivity, and concentration of Cl-, NO3-, SO4(2-) F-, Na+, Ca2+, Mg2+, K+, NH4+, Cu, Zn, Fe, Mn, Pb, Ni, Cd, Co, and Cr. SO2 and NO2 distribution over the time were also evaluated. Results have showed that pH < 5.6 are found mostly at Candiota airport (85%), followed by Aceguá (72%) and Três Lagoas (65%). Enrichment Factor of the studied ions in wet deposition revealed higher Ca2+ and SO4(2-) enrichment in Três Lagoas. Factor Analysis applied to metals and major ions allowed identifying the major sources. While Cl-, Na+, Mg2+ are of marine origin, SO4(2-), NO3-, NH4+ ,F- come from anthropogenic sources. Except for Fe and Mn originating from the soil dust, the metals studied showed to have anthropogenic influence The average SO2 and NO2 concentration, as well as SO4(2-) and NO3- in wet precipitation in the Candiota region showed higher concentrations during the warmer months.

A new world natural vegetation map for global change studies

We developed a new world natural vegetation map at 1 degree horizontal resolution for use in global climate models. We used the Dorman and Sellers vegetation classification with inclusion of a new biome: tropical seasonal forest, which refers to both deciduous and semi-deciduous tropical forests. SSiB biogeophysical parameters values for this new biome type are presented. Under this new vegetation classification we obtained a consensus map between two global natural vegetation maps widely used in climate studies. We found that these two maps assign different biomes in ca. 1/3 of the continental grid points. To obtain a new global natural vegetation map, non-consensus areas were filled according to regional consensus based on more than 100 regional maps available on the internet. To minimize the risk of using poor quality information, the regional maps were obtained from reliable internet sources, and the filling procedure was based on the consensus among several regional maps obtained from independent sources. The new map was designed to reproduce accurately both the large-scale distribution of the main vegetation types (as it builds on two reliable global natural vegetation maps) and the regional details (as it is based on the consensus of regional maps).

Two fast screening methods (GC-MS and TLC-ChEI assay) for rapid evaluation of potential anticholinesterasic indole alkaloids in complex mixtures

The pharmacotherapyfor Alzheimer's disease (AD) includes the use of acetylcholinesterase inhibitors (AChEI). Recent investigations for novel AD therapeutic agents from plants suggested that Tabernaemontana genus is a promising source of novel anticholinesterasic indole alkaloids. In this work two fast screening techniques were combined in order to easily identify novel cholinesterase inhibitors (ChEI). Gas chromatography-mass spectrometry (GC-MS) of the less polar alkaloidic fractions obtained from the acid-base extraction of the stalk of T. laeta revealed thirteen monoindole alkaloids, four of them confirmed by co-injection with previously isolated alkaloids. The others were tentatively identified by mass fragmentation analysis. By gas chromatography with flame ionization detection (GC-FID) and using isatin as internal standard, affinisine and voachalotine were determined as major compounds. These fractions and fourteen previously isolated alkaloids, obtained from root bark of T. laeta and T. hystrix were investigated for acetyl (AChE) and butyrylcholinesterase (BuChE) inhibitory activities by the modified Ellman's method in thin layer chromatography(TLC-ChEI). Results showed selective inhibition of the alkaloids heyneanine and Nb-methylvoachalotine for BuChE, and 19-epi-isovoacristine for AChE, whereas olivacine, affinisine, ibogamine, affinine, conodurine and hystrixnineinhibited both enzymes. In addition to confirming that monoterpenoid indole alkaloids can be novel therapeutic agents for AD, this is the first report of the ChEI activity of olivacine, a pyridocarbazole alkaloid.

Identification of the antimicrobial substances produced by Solanum palinacanthum (Solanaceae)

To find out natural antimicrobial agents as alternative in therapeutics and to preserve food, the methanol extract of Solanum palinacanthum aerial parts was submitted to purification steps guided by antibacterial and antifungal assays. As a consequence, the flavonoid rutin and 3,5-dicaffeoylquinic acid were isolated by column chromatographyand high performance liquid chromatography, and identified by mass and nuclear magnetic resonance spectrometry. Minimal inhibitory concentrations (MIC) of the quinic acid derivative against Aeromonas hydrophila, Bacillus subtilis, Staphylococcus aureus and the fungus Aspergillus ochraceus were 250, 1000, 1000 and > 568µg/mL, respectively. Against the same microorganisms, MIC for rutin were 1000, > 1000, > 1000 and 35µg/mL, respectively. Rutinwas very promising for A. ochraceus control, since its MIC against such fungus was close to the one observed for benzalkonium chloride, which is used as a fungicide in Brazil.

Evidences of delayed size recovery in Araucaria angustifolia populations after post-glacial colonization of highlands in Southeastern Brazil

Up to date, little is known about the relationship between historical demography and the current genetic structure of A. Angus As a first effort towards overcoming this lack, microsatellite data scored in six populations and isozyme allele frequencies published for 11 natural stands of this species were analysed in order to assess molecular signatures of populations' demographic history. Signatures of genetic bottlenecks were captured in all analysed populations of southeastern Brazil. Among southern populations, signatures of small effective population size were observed in only three out of 13 populations. Southern populations likely experienced faster recovery of population size after migration onto highlands. Accordingly, current genetic diversity of the southern populations gives evidence of fast population size recovery. In general, demographic history of A. Angusmatches climatic dynamics of southern and southeastern Brazil during the Pleistocene and Holocene. Palynological records and reconstruction of the past climatic dynamics of southeastern and southern Brazil support the hypothesis of different population size recovery dynamics for populations from these regions.

The importance of feeding in the larval development of the ghost shrimp Callichirus major (Decapoda: Callianassidae)

The present study investigated whether the callianassid Callichirus major shows a lecithotrophic behaviour during larval development. Two experiments were carried out. In the first experiment, larvae were subjected to an initial period of feeding, while in the second they were subjected to an initial period of starvation. In Experiment 1, 80% of C. major larvae succeeded moulting to juvenile stage in treatment with larvae fed every day. In the treatments with larvae fed for 1, 2 and 3 days there was total mortality before they reached the megalopal stage. In Experiment 2, zoea larvae showed more resistance when subjected to an initial period of starvation in which larvae starved for 1, 2 and 3 days and had survival rates of 100, 60 and 80%, respectively. But, a delay in the development duration of the zoeal stages was observed. Total mortality was observed for larvae reared in the treatment with entire starvation. The results suggest that zoeal stages of C. major are not lecithotrophic.

Anatomy and ontogeny of the pericarp of Pterodon emarginatus Vogel (Fabaceae, Faboideae), with emphasis on secretory ducts

Discrepant and incomplete interpretations of fruits of Pterodon have been published, especially on the structural interpretation of the pericarp portion that remain attached to the seed upon dispersal. The present work clarified these doubts and analyzed ultrastructural aspects of the Pterodon emarginatus diaspores using light and transmission electron microscopes. Cell divisions are prevalent among the initial phases of development, and the subadaxial and adaxial meristems form the fibrous inner mesocarp and the endocarp composed of multi-seriate epidermis, respectively. At the median mesocarp, numerous secretory ducts differentiate between the lateral bundles, by lytic process. After lysis of the central cells and the formation of the lumen, the ducts show unistratified secretory epithelium with dense cells; oil droplets are observed on the secretory epithelium and the subadjacent tissues. At maturity, the uniseriate exocarp and the outer mesocarp slough off in an irregular fashion, leaving the diaspore composed of a papery and brittle wing linked to a seed chamber that includes the median mesocarp composed of lignified cells, bordering vascular bundles and many secretory ducts whose epithelial cells develop large vacuoles that accumulate oleoresins. The Pterodon emarginatus fruit is a cryptosamara.

Crude ethanol extract from babassu (Orbignya speciosa): cytotoxicity on tumoral and non-tumoral cell lines

Plant-derived substances have been considered as important sources of drugs, including antineoplasic agents. Babassu mesocarp is popularly used in Brazil as a food additive, and in popular medicine against several conditions, such as inflammations, menstrual pains and leukaemia. From babassu Orbignya speciosa (Mart.) Barb. Rodr. [Arecaceae (Palmae)] epicarp/mesocarp, an ethanol extract was prepared and named OSEME, which was tested on the viability,morphology and metabolism of several cell lines, such as the leukaemic cell lines, HL-60, K562 and the latter multidrug resistant counterpart K562-Lucena 1, the human breast cancer cell line MCF-7, the mouse fibroblast cell line 3T3-L1 and fresh human lymphocytes. OSEME promoted a dose-dependent decrease on the viability of all cells. This effect was much more pronounced on the tumoral cell lines than on non-tumoral cells, a phenomenon revealed by the dose of OSEME which promotes half of maximal effect (ID50). The decrease on viability was followed by shrinkage of cells, alteration on their morphology, and a markedly nuclear condensation. Curiously, stimulation of 6-phosphofructokinase activity (6.6-times) was observed on HL-60 cells, treated with OSEME, when compared to control treated with ethanol (vehicle). These results support evidences to suggest OSEME as a promising source of novel antineoplasic agents.

Endophytic colonization of rice (Oryza sativa L.) by the diazotrophic bacterium Burkholderia kururiensis and its ability to enhance plant growth

Burkholderia kururiensis is a diazotrophic bacterium originally isolated from a polluted aquifer environment and presents a high level of similarity with the rice endophyte "B. brasilensis" species. This work assessed the ability of B. kururiensis to endophytically colonize rice plantlets by monitoring different tissues of root-inoculated plants for the presence of bacterial growth in different media, electron microscopy and by 16S rDNA analysis. Observations of roots, stems and leaves of inoculated rice plantlets by electron microscopy revealed B. kururiensis colonization predominantly on root hair zones, demonstrating endophytic colonization primarily through the endodermis, followed by spreading into xylem vessels, a possible pathway leading to aerial parts. Although indifferent for the bacterial growth itself, addition of a nitrogen source was a limiting factor for endophytic colonization. As endophytic colonization was directly associated to an enhanced plant development, production of phytohormone auxin/indole-3-acetic acid by B. kururiensis was assayed with transgenic rice plantlets containing an auxin-responsive reporter (DR5-GUS). Our findings suggest the ability of auxin production by plant-associated B. kururiensis which may have a stimulatory effect on plant development, as evidenced by activation of DR5-GUS. We hereby demonstrate, for the first time, the ability of B. kururiensis to endophytically colonize rice, promoting both plant growth and rice grain yield.

Influence of Palythoa caribaeorum (Anthozoa, Cnidaria) zonation on site-attached reef fishes

This work aimed to test the influence of Palythoa caribeorum, a widely distributed zoanthid in the Atlantic, on site-attached reef fish in a subtropical rocky shore. Density, richness and vertical distribution of reef fish inside (ID) and outside (OD) previously chosen P. caribaeorum dominance patches were compared through stationary visual censuses along three different periods. Fishes were grouped in different trophic guilds to evidence differences in resources uses in both treatments. A complexity index was estimated by the chain link method and percentage covering of benthic organisms was obtained analyzing random points from replicated photo-quadrats. We observed thirty-eight species of fishes, belonging to twenty-five families. Reef fish communities between studied patches were similar,both in terms of species composition and vertical distribution. Considering only the most site-attached fishes, which were the most frequent and abundant species, data showed that ID sustains higher diversity and abundance than OD. Results showed that benthic composition differ significantly among patches whereas complexity remained without differences. Otherwise, results indicated that these areas might play an important role in space limitation, structuring neighboring benthic community and consequently reef fish assemblages.

Superoxide dismutase and lipid hydroperoxides in blood and endometrial tissue of patients with benign, hyperplastic and malignant endometrium

Epidemiological and experimental data point to involvement of oxygen derived radicals in the pathogenesis of gynecological disorders, as well as in cancer development. The objective of the present study was to examine changes in activities and levels of copper/zinc superoxide dismutase (CuZnSOD) and lipid hydroperoxides (LOOH) in blood and endometrial tissue of patients diagnosed with uterine myoma, endometrial polypus, hyperplasia simplex, hyperplasia complex and adenocarcinoma endometrii. The results of our study have shown decreased SOD activities and unchanged SOD protein level in blood of all examined patients in comparison to healthy subjects. Decrease of both SOD activity and level was found in endometrium of patients with hyperplasia simplex, hyperplasia complex and adenocarcinoma in comparison to women with polypus or myoma. LOOH level was elevated in both tissues of patients with hyperplasiaor adenocarcinoma in comparison to healthy subjects or patients with benign diagnosis. Our findings suggest that the decrease in SOD activity and level, as well as the increase in LOOH level, in patients with gynecological disorders, render these patients more susceptible to oxidative damage caused by reactive oxygen species (ROS). An imbalance in ROS formation and SOD level may be important in the pathogenesis and/or perpetuation of tissue damage in gynecological patients. Since evidence suggests that SOD may be a therapy target for cancer treatment, our findings provide a basis for further research and options for clinical applications.
